Agar germination or ?

Hey
First time doing agar.
Did a zigzag of MS on DFA on 3 plates.
I'm seeing spots growing from the streaks. several about the size of a dime. It doesn't look like mycelium. Just frosted section. Id take a pic but it wont show up with the glare etc.
If I look real close on the outer sections of growth it looks like stringy fuzz but its way to thin for me to believe its mycelium. Its the only growth on the plates though.
Is it possible to have just spread contams on 3 plates?

Sometimes the first signs of growth don’t show up very strong. You got to wait a bit for the thicker stuff to start coming in. How long ago did you streak your plates, and what agar recipe are you using?

1 week.
Dog food agar
10g dog food
10g agar
500ml water.

Its seems extremely "Stiff"

I’ve never used dog food agar before. A week for germinating spores sounds about right. I’d just give it a little more time and see. Maybe post pics back in a day or two, once you can get a good pic of what you’re talking about. It can be challenging diagnosing a issue without a picture.

I use 8g agar to 8g brf for 400ml of water. There’s many ways of doing it though that people have success with, or not so much.

Maybe someone else can chime in. Like I said though, it will be easier to figure out once you can get a picture of the growth. Let it grow more. Patience, IME.

Looks like bacteria to my eye. I normally just use the smallest amount of spores in the middle of my plate to lower my risk of spreading contams by streaking. That’s just me though, plenty of people streak.

Can’t really tell to well what that is for sure. My first guess would be bacteria. There’s still a chance myc will grow though and you can transfer clean growth to a new plate. I’m still learning the ins and outs of agar myself.

I'd say it's very thin mycellium. The edges have kind of a hairy structure like mycellium. Bacteria often has sharp edges like a drop of water, not always though. I would wait a few more days to see if it gets thicker.

When I’m doubt, wait some more :wink:

Looks like it’s getting thicker, so that’s a good sign.

Give em’ some more time to grow out :thumbup:

Maybe try starting some new plates in the mean time with different nutrients and agar content. Test the waters as they say.

Easier to see sometimes looking from the back, but your agar is murky and colored.  I like mine crystal clear.  Seems fine to me,  I dislike loop streak.  I have great success with a single syringe drop. Dead center. Try not to let it run around the plate before it absorbs.

Gonna toss another vote in the "single drop of MS" box; every time I've streaked, I've been really unhappy with what I had to transfer. Plopping it in the middle usually results in clearer growth patterns for me. (But I could also just suck at streaking. :grin:)

The dogfood agar has lots of sediment. and I added food coloring to it of course.

That's exactly why I didn't try DFA again; hated how gritty it came out. I'm a big fan of malt extract agar. Good clarity, tried and true recipe.

10 grams light malt extract
9 grams agar powder
500 mL distilled water

Prep just like you would with the dog food agar, except now you don't have to keep a coffee grinder full of dog food dust sitting around.

That's fair; this is just the recipe I started with and kind of stuck with. When I was in a dryer climate, I appreciated the slightly wet quality of it and I've just stuck with it since.
