Relatively new, enjoying my recent success and trying to build upon it.
So I started growing about 6 months ago, with a failed attempt at brf and a failed try with spore syringe to honey water LC...bad idea I know now. Since then I have had 6 successful grows with spore syringe to brf cakes, all with several aggressive flushes, 100% take on 10-20 jars each time, 100% without contamination, using B+ and GT spore syringes from Shroomery trusted vendors. Ive spent what must be hundreds of hours sifting through the threads here and truly appreciate the education, as well as the lessons my mushrooms teach me. Now I had 4 fully colonized agar plates, made myself with teks found here, of which I used one to make a few LC jars, and one to try wedge transfer to brf cakes and to make an LI syringe using teks I found here that seemed well-liked. Unfortunately, my LC failed again (3 jars with the agar wedges, 1 with a fresh cut from inside a fresh picked mushroom) I wish I could get a viable non-contaminated LC but idk what i'm doing wrong...2 of the jars were ELME only, 2 were ELME/dextrose, both at 4% total:water ratio... all 4 jars looked too dark despite coffee filtering twice then PC'ing again, and had barely any growth around the agar wedges/shroom in the LC jars, and when I tried using them to inoculate cakes had no growth at all in 3 weeks...only let them sit that long bc I have enough fully colonized brf cakes and flushes going in my 2 SGFC's that my hands are full enough atm. The LI i made with the myc from the agar also didn't take after immediately injecting to sterile brf cakes.
So I still have 2 fully colonized agar plates from my more aggressive growing mushrooms, and 4 more plates I am watching expand (beautiful rhizomorphs growth) now which I made using small wedge transfers from the other 2 plates. I have a couple bags I bought from outgrow, which I wanna try inoculating: a manure bag and a rye bag (decided to try first time with these items from a pre-sterilized, pre packaged source, though I make my brf cakes on my own) Have 1 jar of rye I made and sterilized in the PC a week ago...only reason not inoculated is bc I didn't wanna inoc that with spore syringes, I wanna use live myc. I heard properly prepped rye can sit without inoculation for a while? And I have a ton of 1/2 pint jars I can make more brf cakes in. My next step is to inoculate something, anything, with live mycelium from my agar plates. So far I have not been successful with making a viable LC, LI, or even wedge transfer to brf (yes without the dry verm layer, tyvek filter in lid instead) The wedges transferred to new agar plates are growing great. No signs of the white bacteria I hear people mistake for mycelium...just perfect rhizomorphs growth similar to the initial growth in my brf cakes. So I am convinced my agar plates are good. Shroomery is full of agar making/pouring teks, but disproportionately low on what people do with their agar once its there, grown, stored in the fridge, waiting for use..
Also, I have access to basic medical supplies like hospital grade sterile water, sterile syringes/needles. I have my PC, I have a SAB, plenty of 70% iso alc. . .
TLDR:
I have agar plates growing, (lets give the benefit of the doubt to my babies), uncontaminated mycelium, 1 even pinning in the petri dish, IN the fridge which is very cold!
My questions are: 1) What should I do with them to inoculate brf cakes (1/2 pint PF tek jars)? 2) What should I do with them to create a viable LC 3) Does anybody prefer making their LC's using fresh mushrooms, because I have more of those popping up all the time right now 4) Do any of you make LC's or LI's using shredded fully colonized/consolidated brf cakes? 5) On the side, I want to get the rye and manure bags I bought started. They are presealed, sterile, with injection ports and air exchange ports. What is best to inoculate them with given what I have available (agar, colonized brf cakes, fresh shrooms) 6) If I have to make one of those DIY jar-with-blender-blade things that can be sterilized in the PC, what is the simplest one to make involving the least $ and bob-the-builder skillz? lol
