Licky711 first grow the journey begins

So last night I made a sab for the first timeout of a 100 liter plastic container. I pulled my Petri dishes from the fridge they were premade lab quality potato dextrose agar vacuum sealed so didn’t autoclave them. Sanitized the sab with Lysol and rubbing alcohol put my gloves on pulled my Golden Teacher spore print out scraped it with the inoculation loop and sneed it over two Petri dishes pulled the Parafilm out and wrapped them the best I could and put them on the top shelf. The temp is about 25-27 Celsius humidity is kinda low only 35-40% I have to run a space heater to keep the room warm.

So a few questions 1. how long should it take for me to see in there is in contamination?
2. What temperature should I keep it at?
3. How’s the humidity ? And what should it be at?

I’m going to do 2 b+ , 2 Texas/PE6, 2 treasure coast, Panaeolus Cyanescens Jamaica, 2 Brazilian cubs. Today today is a good day. Also I just ordered a humidifier and I’m looking for a flow hood to order since the last manufacturer wasn’t able to send it.

So I just picked up a uv disinfectant light for the grow room and a humidifier. Does anyone have experience with using a uv light in there mushroom room any tips or tricks to keep a clean environment?

Honestly, they're both unneeded. A germicidal lamp only works in a proper clean room where the surfaces are unable to harbour any contaminants. Any carpet or furniture in the room can harbour contaminants and shield them from the light. A germicidal lamp will only create harmful concentrations of ozone in the room you'll be working in.

More importantly: as long as you practice proper sterile technique in a SAB, you could be working in an ebola-infested crack den and still have good results.

The humidity in the room doesn't matter. If you're going to do a monotub (it sounds like you are), then the humidity in the tub itself isn't even that important. Just keep the substrate glistening so that water continually evaporates from the surface.

My agar hasn’t grown anything in three day. What should I do? How long does it take to see growth?

Depends how old the print is/how dry the spores are. Could be anything from a couple of days to a couple of weeks:shrug:

They were pretty fresh spores from a sponsor from this site. The spores were almost wet lol

Sadturkey it was premade agar dishes I bought from a lab. I still haven’t made my own dishes.

Slow down and take a deep breath. You don't need to run out and buy fancy gadgets.  You don't need anything other than some of the simple methods listed at the top of the cultivation forum and good sterile technique.  It's very exciting in the beginning but focus on the basics first.  Learn how to make your own agar dishes, get your sab techniques down, learn how to do a simple transfer with your plates. That will serve you better than over complicating things.  Good luck and be patient - these things take time.

you might want to make you own agar. you can buy agar-agar powder and then go to a home brewery store/beer store and ask for "light malt extract". LME is crystal clear. the reason you want LME is so you can make agar weaker or stronger depending on what you need for your culture.

but since you already have premade just use that and if its too strong dilute it with more water. when i say strong i mean that the growth on the plate has a tendency to grow very dense and "tomentose". when you use "weak" agar concentration you get ropey "rhizomorphic" growth that is easy to distinguish from contaminants that hitch a ride.

So it’s been 8 days only see some light small growth. and on one plate a small green or brown satellite colony I’ll add pictures in the morning

I don’t know what to think about these agar plates is it doing ok ? It’s been 8 days and they haven’t grown much? What should I do now ? It’s my first time doing any of this.
