Can you blast peroxide agar with UV or sterile catalyste to destroy peroxide?

in light of recent contamination ive considered peroxide agar, also i have some absolutely filthy AA+ cultures which i cant get going off the only spore print i have left of them.

I got to thinking, if peroxide agar kills spores on contact, doesnt that mean that a little while after the dish is sealed, the only living things left should be the mycelium. If peroxide is broken down by UV, wouldnt it be possible therefore to use UV to decompose all the peroxide in the sealed dish, which then generates a nice healthy environment for the mycelium to colonize and live on for the long term, as far as long term goes for agar anyway. Or at least to prevent mutations or stunted growth, or other damages. Thats all im really concerned about.

Another alternative option would be to introduce a soluble dissolved sterile catalyst to the dish during the transfer, which would over time break down the peroxide. achieving the same result. Or perhaps simply tainting the peroxide or agar with a catalyst initially so that the peroxide has a fairly low life span in the agar, destroying it shortly after it serves its purpose.


I do realize peroxide is not a popular thing around here and is not needed when you simply improve your sterile tek, and i am working on that, but, for the time being, is this a viable option? UV, catalyst or catalyst in the agar?

I have had some luck flipping the transfer "mycelium side" down. It allows the mycelium a chance to leap out faster than the bacteria that is trapped between 2 layers of agar. I have started to make all my dishes more shallow to make this easier. I think this is becoming a standard procedure for me. Doesn't hurt anything at all, and doesn't take any longer. Trick is to have thin agar.

Other wise your better off getting some antibiotics. Peroxide does some damage to mycelium as well.

no, neither UV nor peroxide is a viable alternative to sterile technique

whatever you are reading about peroxide agar in is VERY old, peroxide is basically useless in our hobby

well timed transfers and streaking technique is ALL you need to get clean cultures. just figure out what you are doing wrong. the more info you give use the more we can help

I have heard that UV causes peroxide to break down, not form, the purpose would be to get rid of the peroxide once it has served its purpose of preventing spores from germinating. Also this is not an alternative, it is supplementary. Gloves for example are also supplementary, you can argue that they are unneccesary and its possible to get by just fine without them, but its still better with them. I do however recognize how limited the application of UV is.


Anyway, the 1st and second batch of petri dishes i did since i began using my SAB on a flat desk in the house instead of in the filthy shed, turned out great, no external contaminations, although, unfortunately none of the clean spores ever germinated, others it turned out were contaminated and so, green mold began to grow before the spores which still have done nothing after 2 weeks, and this was on normal MEA agar ill just note.


Last time and the time before (3rd and 4th since comming inside) however, still doing everything the same, i started getting massive contaminations of everything. I dont know if the source was from one of the contaminated cultures i tried cleaning, or something i simply did wrong.
the previous time i decided to ditch the sleeves on my SAB and just wear gloves, but still, the same contamination happened again, so, it wasnt suction or anything like that.

i used to have constantly inconsistent dishes when i used the lid for the SAB floor, it had an air gap so, was allowing air to move freely in/out, and i now thought i had it down with just putting it on a flat surface, but again, the same issues. Dont understand how some times its perfect and other times not. Thats why im building a flowhood. But in the mean time, i want to try using peroxide agar and minimize the damage it causes.


I managed to isolate some especially nice creeper mycelium, and also finally managed to get golden teacher growing from an old contaminated syringe, i dont have any spores left to try again with though, both are contaminated and both only have very small growths, so, i only have a limited number of chances with them, they are the real reason i even considered it at all.

The peroxide would degrade soon anyways.
And is useless

If you have no luck with a sab a flowhood isn't s magic solution just because it's fancier

try to find 1 person who knows what they are talking about and uses peroxide agar in the current year... there is a reason no one does

you dont need anything to supplement clean technique. it isnt a matter of damage, its pointless

On IRC i spoke with someone who occasionally does when cleaning up cultures. They are the ones who recommended it.

The issue still remains that i have dishes of peroxide agar, how should i go about getting rid of the peroxide, without harming the mycelium, would UV/sunlight work? or would that be ineffective and cause more harm than neccesary to the mycelium?

Uv b light is evil. I thought solve some of my cleaning problems but it burn the shut of my skin that it came in contact with. Peroxide with degrade within a few days without the light

i dont plan on using it again anyway, probably wont need to anyhow if the flowhood works.

So, whats the game plan, should i use a UVC lamp, which i already have, or sunlight? and then, should i just stick something on the dishes to shield the mycelium? or is that not neccesary with direct sun, and how much/long? If you happen to know how long i should before the h2o2 levels drop enough that it wont bother the myc.
i used 3ml of 6% H2O2 in 300ml of potato dextrose yeast agar

I do appreciate your advice thus far.

added the peroxide shortly before pouring it while it was still runny enough, but not so hot that it would cause it to decompose

o.0 You know peroxide turns to water from like even the smallest amounts of light, thus the hardcore dark bottles. Shit is probably water by now anyways, so don't sweat it too much.

As others have said, work on techniques.

I've seen 0 evidence about tea actually being useful for mycology. I've seen some trying to exploit the small antibacterial qualities of black tea however tea is a bacteria magnet and it's pressure cooked/autoclaved probably destroys all of its slight antibacterial properties

there is just no need for them, like at all... the sooner you realize that your agar game is not going to improve by adding some special ingredient to your agar, and that the point of agar is controlled growth in 2 dimensions, the sooner you can start focusing on things that will actually affect your outcomes

I've used activated charcoal before, It makes your plates look cool. that's all I noticed. I would be extremely surprised if it inhibited bacteria though

BL sent me some charcoal to try and dog food for dog food agar :lol:
DFA works in a pinch but otherwise it's about as gross to work with as BRF agar.

i have some black food coloring that looks cool :smile:

id be scared to work with dfa... too much meat and stuff in there, i bet some NASTY pathogens can grow on DFA... i prefer to use as selective of a medium as possible... kinda like extracting thc
