peroxide instead of sterilization?
    #2328717 - 02/13/04 12:30 AM (13 years, 2 months ago)

I found a link to a book by a PhD who claims that you can use hydrogen peroxide to eliminate the use of pasteurization/sterilization techniques. the book comes in two volumes for a total of about $30. is there any truth to these claims?

the man's name is R.R. Wayne, Ph.D. and the site is www.mycomasters.com

is any of this true?

Well, if you are talking about the h2o2 manuals,I'm pretty sure its clear that h2o2 only kills spores. The need for pasteurization/sterilization is,as far I know, still in force.

buy the book and inform the rest of use what is says and if it works. not worth my money, mmgg is free and time tested.

The theory is that once spores have germinated and been exposed to h202, that it doesn't affect the growth(much) anymore. So, you could introduce the germinated mycelium into h2o2 rich substrate, and it would be able to colonize it.. while the h202 would destroy any invading spores, keeping them from germinating.

Hey, I came across this old thread in the process of doing a search to see if a h202 sterilization tek I thought of was novel...I guess not. Anyways, I'm wondering if anyone knows the specifics of mycelial resistance to h202 and just how much bacteria the 3% h202 solutions you buy in the stores kill.

My idea was rather than using h202 in g2g transfers for instance, simply use h202 in lieu of water in the PF Tek. Assuming the h202 kills a sufficient amount of bacteria, one would only have to wait until the h202 decomposed (upon exposure to light, heat etc.) before innoculating and you would have a sterile jar without having to deal with steaming/pressure cooking. Anyone have experience with this or have heard of experiments using a similar procedure?

If not I'm willing to give it a shot (no pun intended).

H202 doesn't kill bacterial contaminants, or competetor fungi, only spores. It's a useful safety net for mycological work, but it cannot practicly replace heat sterilisation.

here is volumes 1 & 2 of Growing Mushrooms the Easy Way by R. Rush Wayne, Ph.D.

http://www.nansnook.com/~archives/tek/Volume1.html

h2o2 is great, but it won't do what you're talking about... (you can use it very effectively to guard against airborne infection after innoculation, but it doesn't sterilise anything)

Ahh, a very useful link...thanks

I have a fractional sterilization experiment running at the moment (http://www.shroomery.org/forums/showflat.php?Cat=&Number=2566301&page=0&view=collapsed&sb=5&o=&fpart=1)
and as I am nearly out of spore solution, I was planning on using some partially colonized BRF cakes as spawn. Would a sufficiently sterile procedure just be to remove a small piece of the colonized cake, clean it of uncolonized substrate and dunk in h202 before dropping it into the grain?
