4 inoc site will give you a MUCH higher chance of contamination, but will also make colonization faster.

1 inoc site will make it colonize VERY slow, i would only recommend it if you don't care how long it takes and if you aren't sure if you can sterilize everything very well.

if you do decide to use just one inoc site, put at least a whole cc in there and try to spread the spore solution around by tilting it.

I would definitely recommend against innoculating only one site. Colinization will take much longer. While there may be less chance of introducing a contaminate in the innoculation stage, slow colonization increases the chance of contamination.

Drill only one hole in the center of your lid & then insert the needle facing left...inoculate...and then take it out and insert the needle in the same hole facing right...inoculate again... Now you have two inoculation points on either side of the jar using only one hole.
Just a thought...

exploding boy has a good idea. I've always just used two holes and this has worked fine for me. You can always use more innoculate which will speed colonization time.

I don't think the number of holes matters. Just as mentioned previously, innoculate in several different directions. It would disperse the spores over a wider area and make colonization quicker than having one mass of spores in one spot.

Yea, good points. What I treid recently was squirting a bit to the left, the center and then the right in EACH of the four holes. That's 12 innoculation site, which may take more innoculant than you want, but they colonize pretty dman fast.
