Not if isolated from a clone, but if from multispore, yes.

Not common, but it does happen.

Culture on agar growing slowly and cottony is typically monokaryotic and incapable of producing fruits unless mated with a compatible monokaryon .

Sometimes partial mating compatibility between monokaryons (common-A and common-B pairings) produces dikaryons unable to form fruitbodies. These can often be distinguished from sexually competent dikaryons under a microscope, because they show abnormal cytological features - absence of clamp connections, incomplete septation, etc. Such strains are most likely to arise when spores from the same fruitbody are crossed, because these spores will have the highest proportion of common mating alleles.

I have used agar for years. I have never seen any rhizomorphic growth on a multi-spore innculation on agar. I have also never seen a strain that didn't show exceptional rhizomorphic properties close to fruiting or a strain that didn't fruit like crazy. I guess I've been lucky, but after probably 100 agar plates, you'd think I'd have seen this mystical "non-fruiting" strain that everyone warns against.

I was really thinking about matings between partially incompatible mycelia derived from single spores.

With a multispore inoculation, I can see how you might get infertility problems were you to isolate very small patches of mycelium immediately after germination, but does anyone actually do this?

In a more mature multispore-derived mycelium, fully compatible pairings of nuclei will have come to dominate the network. Therefore any region you isolate is likely to be capable of fruiting.

I am in complete agreement. I think that ps cubensis is such a ready-to-fruit kind of mycelia, it's hard to ge a non-fruiting isolate. Perhaps with other species that are harder to fruit, such as the ones Stamets talks about, it is possible to have a non-fruiting strain. I don't think that nature would allow for amycelia that could grow, but not produce fruitbodies, no purpose.

You've never heard of infertility? Just because something can't reproduce has nothing to do with its ability to survive. Sure it won't be able to pass on its genes and continue its genetic line but that specific specimen still has purpose and will still grow, continuing to breakdown its food just as any other mycelium of its species.
