Other than the fact that I dreamed last night that every plate I had inoculated recently got contaminated (🤣🤣🤣), it's actually going really well.

The inspection hatch covers were pretty plug and play. I basically put the bezel on the SAB and traced it with a sharpie and then cut The circles out with a Dremel (wear your safety glasses!). I cut them small and gradually adjusted for a tight fit, so no silicone necessary. The one on the right actually doesn't fit perfectly and I've been meaning to adjust it a little bit. Silicone would work but so would screws. Other than not having a permanent home for my SAB, I've been really happy with it.

After much debate with myself, I gave up on building an induction sterilizer and got an infrared one instead.

I agree. I use an induction coil right now but both of these devices evoke the same question in my head.. Why place it outside the SAB?  Constant moving of your arms and hands in and out of the SAB, it seems to me, will only increase the turbulent airflow and kick stuff up that can land on your plates or in your jars. There is no open flame so there shouldn't be an issue with explosion risk. Also, I don't spray the inside of my SAB with ISO (although I do keep a jar of it inside the SAB to drop my scalpel into in between uses)... So what is the downside of keeping the sterilizer in the box versus outside?

Heat source generates way more air currents than slowly moving your hands in and out. Hot air rises.

This ^^

Also the thing itself gets pretty hot, as well as the air near it. This was the biggest and most powerful I could get - 380W (though my UPS said it's drawing more like 400W). It's quoted up to 850C, and while I don't have any means to confirm it, I can attest that it gets really hot. I don't think it'd be possible to work with it in there even if you wanted to.

I put the wider nichrome tape in, I'm going to have to rig something up to shorten it a bit, the ends glow if I try to crank it up to seal over sawdust.

ooo! that’s next level. The elevation adjustment is also cool. That would solve a “sorta” problem that I have had with trying to get a solid bag seal with the one I’ve got, with bags laying on their side…always an uncomfortable operation.

Hey guys,
I picked up something similar on an impulse buy from a UK supplier but they are shitty with their product description and communication and I didn't really think about it when I ordered it.... Is this lid garbage? How do I use it?  The silicone tube has a luer fitting at the end, and I have a cap for it. There is a stopper on the  outer tube. Why is the outer tube there at all, and why is it so long?  I suspect they used a second piece of tubing because securely attaching a luer fitting directly to the plastic lid is fiddly?

I was thinking this could be used without a SAB but this doesn't seem to be the case?

I am now not sure i want to do LC at all since the contamination can be invisible and only become apparent once it is in my grains....

I was thinking this could be used without a SAB but this doesn't seem to be the case. I could attach a syringe and sterilise it with the syringe attached, but it seeems that the syringe has to be removed in a SAB and can only later be re-used to make further withdrawals only inside a SAB?







Might be usable.
The tube on the inside should reach the bottom of the jar. The tube coming out the top seems a bit longer than necessary to me.
I'd prefer a cap to put on that luer connector between uses.
It seems like that should be functional.

Thanks. I guess it does need be used inside a SAB, as contamination could enter when attaching the syringe, right?

Disclaimer: I don't use anything like this. But as for sterility, you don't need to leave a syringe attached, you sterilize with the cap on. When you're ready to draw out LC, the bit the cap covers is still sterile. When you sterilize a syringe, you can also cap it, or just keep it in the autoclave bag or jar or whatever you sterilized it in until it's time to use it. Then the sterile end of the syringe is attached to the sterile end of the tubing and you can safely draw out the LC.

As long as you inoculate the LC with clean agar and your technique is good, LC is pretty great. Using a much lower nutrient ratio than you have probably read about (like 0.2% malt to water) really helps keep it super clear, so that any bacteria will be really obvious with its turbidity (cloudiness). Mold is harder to detect in LC, but usually pretty easy to spot on a prospective donor plate.

Thank you. Yeah, brain fart about sterilising with syringe atatched. scratch that, lol.  I bought this because I had trouble getting mycelium out using a needle in the past and thinking I can use larger jars with this, but I guess larger jars aren't really necessary.
