Sectoring

Sorry abut the poor picture, I havent sorted out the condensation problem yet.

Can you tell me what part would you sector an how many would you take?

This is my 3rd transfer and I think im getting close to my first isolate.

This is the bit where i get confused,  how many isolates should I be going for? 

Feedback please....:laugh:

Ive now transferred from 6, 9 an 12 oclock to new plates. psilocybe cyan

Im going to go for 3 and see how that goes.  I think this may be the last one but would still like some feedback.

I have done the same with 2 cubensis plates which are a little behind this, maybe 2 more transfers.  Pastys agar tek is good but im struggling with condensation....

When I get my isolate can I then keep transferring to keep it alive. 

I want to try slants but the missus is moaning about the cost of it all...........

Take as many sectors as you can, try them all.

Tell the wife how it's much cheaper to spend $20 on some slants than it is to have to keep starting over. Long-term costs.

Not really, but I also need glasses really bad. It's extra-hard for me to tell shit from pics.

You're too close.  With that species you want multiple genetics, because fruits are difficult to get and a long time coming.  It's a better gamble to go for a mediocre yield over the 6-18 months you'll be waiting than it is to go for the glory and find out a year later that you have nothing.

Inoculate your grain jars from this plate or the one before.  Just as long as you have a clean culture.  :thumbup:

Yes that makes sence.  Ive made a jar from this plate.  What other species is it best to leave as MS?

That's really the case with any woodlover species, since the accepted way to fruit it is to grow some spawn, then inoculate some chips, then put them outside and wait for a year.

I have 5 jars of ms colonising at the moment.  I just wanted to practice my SAB/agar work.

I have some thai cubes that are going through the same process.  Im on T3 and they arent showing isolate signs yet. 

Look out for this thread as I will post them up in 7-10 days.

Can you tell me, what is a decent speed for one plate to colonise?  Am I looking for quicker plates than others as a sign of speedy growth?  Reading that it seems a little obvious but I still want to double check the method. 

Is there anywhere that talks/writes about the fundamentals of isolating/cloning and what my goals are, the amount of plates that I should aim for ect?

Cubensis is so easy to fruit you can go all the way down to an isolate before fruiting, for sure.  It's a gamble that sometimes doesn't pay off with fruits, as seen in pussyfart's old PE thread, but it can work.  A more commonly accepted way to do it is to partially isolate on agar (maybe 1-2 transfers), then do a monotub and clone a nice fruit.  That way you know you have fruiting genetics.  Isolates are a nice thing to have, since you can use them for experimentation, but they're not strictly necessary.  Since sometimes a good culture is better than the sum of its parts, you can damage it by isolating it too far.

Temp changes cause condensation, so stack an empty dish or two on top of it if necessary for insulation.  Place in ziplock bags too as they help to slow down the temp swings which lead to condensation.

Im just getting sent some Portabello, Shitake and oyster spawn.  Should I try to Isolate these?

What is the best way to expand the culture?

they will already be isolates.  expand immediately on a plate, clean it if necessary, and transfer at least one time even if it is clean, and try to slant them ASAP.  the edible LC's i have in the fridge from 4 months ago are still good, but i still have back ups on plates.  i would be angry if something i paid money for died b/c i was careless.
for growing, expand like cubensis.  agar>master grain>g2g (is u want, edibles do not need high spawn ratios like u would want with cubensis)>substrate>fruit

portabello use poo/compost
shiitaki use sawdust/woodchips/fuel pellets, primarily made of hardwood
oyster use ur imagination for that one :lol:  anything probably will work.  i use straw and fuel pellets

there are a couple scattered here and there.  u saw the RR one?  u can put a single grain onto a plate to expand it for safe keeping.  do maybe 1-2 plates of each.  u can spawn with the rest.  mix with 1-2 qts preferred substrate for whatever species it is.  u don't need a super high spawn ratio with edibles like u would usually do with cubes.

With agar im looking for info on speed from 1st transfer to 4-5 down the line.  Just need to double check im doing it right.  My plates are taking 10 days each at least till ready.

Im currently getting the starch yeast agar dextrose ingredients together to give that a go.  Im sure my growth is slow.

more nutritious agar recipes will usually slow growth down, but thats not necessarily a bad thing.  experiment with many mixes and ill find one u like.  10 days isn't too bad, are u talking about from a wedge transfer?  how big is it after ten days?
