no dikaryon in clone = no friuts?
    #1539750 -

Hey shroomers, this is my first post here!
one quick question/problem:
in order to continue a cake that fruited well for future generations I used some mycelium from the centre of the cake to clone it to a culutre jar that i then used to inocculate several other cakes. But now when I tried to fruit these cakes they refuse to or produce deformed fruits! :frown:
Is it because i happened to use monokaryotic mycelium in the cloning, instead of dikayotic? Should i  have used a fruiting body for cloning instead?
Any ideas, what did i do wrong? Thanks in advance to anyone who replies!

Big words.. Bahumbug.. I dont think u can clone from mycelium.. I think its more like picking out teh strongest most agresive mycelium..But im no expert.

Thanks magic and poke.
Poke smot, I've figured that much out, what i'm asking is it possible that i used mycelium that was monokaryotic despite using mycelium from a fruiting cake?
Should u always use a fruit to clone?

I believe that you did a transfer, not a clone.

It is possible to transfer from mycelium but if the mycelium smaple was too small (like less than a quarter inch chunk) it could have been monokaryotic. If the growth from the mycelium was rhizomorphic or sectored (it should look like the edge of a feather) then the sample was probably not monokaryotic and you have other problems. Monokaryotic growth is tometose (looks like cotton) and does not produce fruits.

if you had a cake of good mycelium, then you automaticly had dikaritotic cells and they never go back to monokarions.

your problem is a manifestation of senesence or contamination. this question belongs in the advanced forum. good question
