Sterilizing Substrate In Advance to Innoculation?

Hi, I used to post here a while ago when I 1st tried to grow mushrooms, and failed, my jars got contamed and dried out when at full colonization ><.

Since I had a contam problem I've been thinking a pressure cooker is definitly what I should be using and I just ordered my spores from TH. Know any good internet deals on a nice pressure cooker, or is Wal-Mart best? Anyone have any preferance with spore purchasing places? And has anyone tried the Thailand-Koh Cubensis from TH? Cause that's what I ordered ;-).

Ok, now for my real question. Could I Sterililze my jars like a week to 2-3 days in advance, to check for contam, etc? And how long after sterilizing would they still be good, so when I get my syringe I can just inject the spores and start waiting for mycell growth?

One more question ;-). Does the bottom of the cake need to colonize before giving them indirect sunlight to trigger the fruiting stage? Or is it ok, if the jar is all white cept for the tippity top?

Thanks so much for you time, I've been reading alot of the beginner faqs and these are the few questions I've came up with.

I dont see a problem with doing that. I did a run where I had 3 extra jars that had been sterilized with medium in it. The lid was on tight, and the ijection holes were sealed with tape. When I took the lids off 1 month later (needed jars for big run), they were still sterile but felt kida dry. I think that as long as they were sealed as to prevent drying out they should be good to go.

Even without a pressure cookeryou can still sterilize. Just have them in a pot with a lid for about 45 minutes,while the water is boiling. The pressure does not kill the bacteria, the heat does.

Ummm, are you refering to the bottom of the jar when letting veg or the bottom when you flip the cake out?

IF you want a good deal on a PC you should probably not look at internet providers, first most places wont ship a PC for free, in other words ur spending way much more then you need to.  Honestly i looked around for a while to find the best deal on a PC but only found a $3-4 difference so its not really worth the gas and time to try and find the absolute cheapest...go with any -MART for one.

If you properly sterilize your jars they can last a couple days to a week.  It is better to inoculate a day after though, as i doubt you will see contams develop before you inoculate

Lastly the whole jar should be white except the tippy top, which is dry verm. You will find that the bottom usually is the last to turn white anyway...(except for my uncle's ????)

As far as TK-cubs i dont kno...hope the rest helps?! :smile:

i got a brand new electric 8qt PC on ebay for...i think the ending bid was 45$ plus 11$ shipping. Also got jars 22 for 20$ including shipping and handling, I find that ebay is the best place to get stuff for this type of thing.  But if u do go shopping on ebay, Be smart. Read the entire thing and ask questions about the product. :smile:

I was very pleased with the KS syringe I purchased from THE.

I bought a 20.5 qt All American on E-bay for $76 including shipping. Buy quality IMO if you can afford it.

If PCing your jars...don't worry about contams from the jars. Be carefull in your inoculation procedure.

Thanks for the reply's, I'm on a tight bugjet, found a 4-quart PC for 18 bucks at wal-mart, and theres an 8 quart one for 40... probly gonna have to settle on the little 4-quarter.

Ummm... oh yea.  on the box it lists the pounds of pressure, it doesn't say PSI, so is 15 pounds of pressure 15 psi?  or how does that work.. cause I think the 4-quart one only goes to 10 pounds of pressure, heh... kinda confusing...

I'm also having trouble with a terranium, I know a cheap 10-gallon aquarium will work, but I'm also trying to think about how I'm going to keep it humidified.... hmm... well I suppose I could just have some sort of lid and drape a think of plastic rap down in there and just spray the wrap to humidify it... the terraniums in the faqs aren't that detailed... I gues is the word, as to how they open and how exactly to use them... hehe, anyone have any detailed plans or anything?

First off it's terrarium. When I first started out I was in the same boat: no cash. I would say that if you're going to go for a ten-gal. terrarium for fruiting and money is a big issue, then buy a bag a perlite for humidification (read the humidity techs.) and strap a couple a sheets of Saran Wrap over the top for an easy, but supprisingly effective lid. Smear a little water over the top of your aquarium before placing the Saran Wrap, this will make it stick like glue. Fan daily, once is good, twice is better. Good luck bro. I think you mean at the bottom of the jar. If this is so, then it is OK to give them direct sunlight. Once pins start to form the rest should be done. If there is only a small patch left to colonize you can also flip the jars to allow for some limited gas exchange. Good luck.....and keep it fictional.

PSI stands for pounds per square inch. If the pressure cooker only goes to 10 pounds you will still be ok. The pressure in the pot only affects the time for water to reach boiling point. Water boils quicker at a higher pressure, hence the use of pressure cookers. If you are on a budget, bypass the need fopr a Pressure cooker, you can still sterilize the jars with a simple pot and a lid. Put water in the pot, put on lid, put your stove on high until you see steam coming from the lid, then put on medium heat for 30 minutes. All will be sterile when done. Good luck.

Not true. Water boils at 212*f at sea level
210*f at 1,025 ft
208*f at 2,063 ft
206*f at 3,115 ft
204*f at 4,169 ft
202*f at 5,225 ft
200*f at 6,304 ft
197*f at 7,381 ft
196*f at 8,481 ft
195*f at 9,031 ft
Therefore the relationship is that as pressure increases so does temprature.

Revrend Stamen's good book says " Note that the effect achieved from sterilizing at 60 minutes at 15 psi is the same as that from 30 minutes at 30 psi.Hence a doulbling of pressure reduces sterization time by one half. "
If you are trying to sterilize, your sterilizing temps will never get over the boiling point of water (212*).Many harmfull bacteria die at 115*-175* ...but thats just the vegetative outside(parent) cell. The tough endospore inside may still survive and contaminate your jars . Grains especially have ALOT of bacterial endospores hence the reason for soaking. Soaking softents of the outer shell of the endospoere and then kill it in the 250*f heat of your "pressure cooker".

I sterilized 6 jars a month in advance for innoc and did 6 a day or 2 before I got my spores....they all are performing the same....if you sterilize properly they will stay good for years apparently...btw I didn't poke holes before hand...so they were vacuum sealed...that prolly makes a diff...but I steamed them didn't use a pc

Direct sunlight will likely cook the mycelium, depending on the season and your geographic location. It's better to use diffused or indirect sunlight.
