please help me. my problem is about spore suspension.

Are you asking how to make a spore syringe?

The only thing spores can be "suspended" in is distilled water.
Normal tap water is impure and will cause the spores to germinate.

A spore syringe is a spore suspension.
Or you can have spores in a jar to suck syringes out of.

Simply take sterile distilled water, and under sterile conditions, add spores from a sterile spore print.

And if everything is STERILE, you will have a spore suspension of whatever spores you want, and they'll last years with proper storage.

If you aren't sterile, every syringe will grow bad things, ruining all your efforts until you weed out the bad syringes.

A suspension of spores or of
mycelial fragments is prepared simply by adding
a few ml of sterile saline to a culture tube containing
the fungus to be studied. The surface of
the tube is then rubbed briskly with a needle so
that surface spores or hyphal fragments are dislodged
into the saline solution. The suspension
need not be homogeneous. With a 1 ml pipette,
a single drop of the suspension is placed on the
surface of each agar cylinder.

thats what im trying to accomplish. but the problem in this method is that the fungus culture is grown in a test tube while my fungus culture is grown in a petri dish. so do i have to subculture my fungi into an agar slant or test tube slant?. or i can just put a few ml of distilled water into a sterile test tube then inoculate a small fragment of my pure culture from the petri dish into the test tube.  then shake it and use a pipette? thnk you very much

just use a sterile loop to remove a few fragments of mycelium from the plate and add to the saline solution

According to RR's 'Let's Grow Mushrooms' DVD, you can add 2 drops of sex lube x 100 ml of distilled water before sterilization to help keep the spores in suspension so they won't stick to the walls of the syringe.

NOT saline! Salt will kill your fungus.
Sterile water.

i assume you're preparing microscope slides. from that:

what you will be observing should be grown in the petri dishes and then ,if possible, you take a slice to look under the microscope, or you can rub a loop in the mycelium and put the fragments collected in a saline solution, mix well, pipette in the slide to look.

for spores:

get the loop, rub a spore print, add to saline solution, shake, pipette to slide.

for spore syringes to innoc substrate:

collect a few spores in a small glass container, add water(tap water will do just fine as well as RO/DI water), suck in a sterile syringe.

for mycelium syringes to innoc substrate:

break a small piece from the agar, add to glass container, add water(tap or RO/DI, but sterile), make a slurry with the myc(use the loop and rub the agar piece under the water to dislodge mycelium strands) then suck in a sterile syringe.

for liquid cultures:

add a small piece of mycelium or spore solution to a glass jar containing a nutrient solution(you can make the same solution you use for the agar, only omit the gelling agent), wait for it to grow, suck in syringe.

therefore:

for observation no incubation times are required, for inoculation of substrates it may be used but is not necessary with small ammounts.
