PDA bacteria

Last week I inoculated several pints of rye with a self made TC syringe. At the same time, I also inoculated 2 agar plates with the same syringe. All were sterilized in the same manner and inoculated in the same ghetto glovebox.

The rye jars are now showing healthy mycelial growth, but the two agar plates are growing nothing but bacteria. I can think of two possible explanations for this:

a) The syringe harbored a contam that favors PDA over rye.

b) The PDA is more easily contaminated and was infected during the inoculation process. Is a glovebox insufficient for working with agar?

Your opinions and advice would be appreciated. In case you could not tell this is my first experiment with agar.  :crazy:

i am a newbie with agar, so im not qualified to give advice here .
but i am wondering, how many plates did you make, and did you incubate any before inoculation ?

dirty tools, too early to tell in the rye, more easily contaminated, all could be reasons. You might want to try peroxidated agar, and cardboard.

I made only two plates and did not incubate before inoculation. On my next attempt I will leave one plate uninoculated as a control. Thanks for the link.
