As long as there are aerioles there, it'll pup again sometime.

I just cut them off and it worked fine. As for size, as long as it's large enough to root before it dies, you'll be good to go. I've rooted pups as small as 1.5 inches, but that was an accident and would recommend you get ones a bit larger than that.

1. it can grow more pups so long as there are more aeroles at the base, which there will be.

2. cut them off with a sharp knife right next to the mother plant.
or you could leave a piece on the mother plant that it will grow pups from if you wanted.

In general a lot of cacti have pups that can be removed easily by basically pulling them off with the twisting motion like you've said.  It's easy to do without any tools and works but cutting works too.  I tend to twist off pups from cacti if I can do it without damaging the plant.  It leaves less exposed tissue than a cut and seems more like the natural way it would happen.

I would wait until a pup is at least 3-4 inches or so before removing it.  Going with the minimum size it can be will lead to a very slow start and possible death.

Pups are a rootstock's first attempt to get more sunlight.  Constantly harvesting them means alot more starting over than the plant is used to. 

Waiting untill it throws out another stalk is a much friendlier approach to harvesting/propogation.
