PF Tek Jars Incubation - question about water content & sunlight & duration

Advice sought please ...

BACKGROUND

I followed PF Tek to make 12 jars:
- BRF:Verm:Water 1:3:1
- boil sterlised 1h30

I followed PF Spore Syringe Tek
- store bought Thai P Cube spore print

And inoculated 5 days ago.

QUESTIONS

1.
How much residual water should I see clinging to the inside of my jars ?
- at room/cupboard temperarure ~20-22°C I'm seeing faint droplets, I reckon over 10-15% of my glass
- at window-sill temperarure ~25-27°C I'm seeing clumpier droplets over all of the glass
- no water pooling at the bottom under either circumstance though

Sound normal ?

2.
I've been told that exposure to sunlight is acceptable during this early end of the incubation stage ... so I'm moving the jars out of the cupboard onto the sill for some sunlight warmth for a few hours each day:
- "not a problem during growth but once its colonized it's prone to pinning"

Sound sensible ?

3.
Will the fluctuating water humidity have any adverse effect ?
- what I mean here is that the sunlight draws some of the water out of the verm into the airspaces and onto the glass inside

... and finally ... the 6 million dollar question:

4.
How many days of zero growth can I expect before I conclude that my inoculation has failed to take ?

I've been told that it could take up to 4 weeks given:
- 20-22-24°C incubation temperature average
- spores were a little over a year past their best by date (but well sealed in the original packaging)

To answer you questions, don sit yur jars in direct sunlight, that will heat them up too much and dry stuff out. light doesnt matter during colinisation, you can leave them in the presence of light/

dont incubate any higher than 80F, 75F is just fine and much safer.

your jars should look dry, it should be a nice light fluffy mix, not moist. being a little wet can slow colinisation.

First signs of growth can be anywhere from a 3 days to over a week, my last jars showd at day 8.

if nthings there by 2 weeks, its generaly not a good sign.


aslng as you got spores from 1 of our trusted vendors, you will be good.

humidity doesnt matter, your jars are sealed in there own little climate

If you stored the syringe correctly, it will outlast prints. which means keeping them in a clean ziplock bag in the fridge, peple hae used spore syringes that old and they worked. Less spores may will be viable, but it still shouldbe good, may take a little longer to see growth.

Spore print was bought in Amsterdam on an overseas trip a few years back. It had a best by 12/2007 date on the base. The print was on a gel, inside a plastic container, inside a plastic bag, inside another plastic container - that's the way I bought it. So sterility was not an issue.

Given the above, I made my own syringe ... 1 hour water boil in syringe, glovebox, flame needle, clean boiled blade ... so sterile advice well followed.

With regard to drying out, I've still got the 3rd aluminium foil layer in place with elastic bands ... so my needle holes are still sealed ... and no evaporation. Once I see some beginnings of mcelium, I'll take off the foil seal, and replace with micro-pore for air exchange.

No sign of any growth at all yet, but I'm happy to wait up to 2 weeks before depressing out - your last 8 day count has given me hope. :thumbup:
