I am fixing to start the PF tek and have a question about what to do after you inoculate the jar. The PF tek says to leave it in indirect sunlight. Some of the other teks have said to leave jars in the dark until the mycelium grow out in the jar . Then put it in indirect sunlight. Do I need to put the jars in the dark are not, maybe I am reading it wrong. Thanks for any help.

Thats the one I am going by. It says to leave them in indirect sunlight. It does'nt say to put them in the dark. At least that is the way I am reading it.

ok. Here is the direct quote from the PF site. "Place the jars in a safe place OUT of direct sunlight." ( I did the cap emphasis). So I would take that as not in direct sunlight. HOWEVER, I do know that people have had them under lights 24/7 for the heat and not had any problems at all. But if you think of it that process in nature is happening underground. You want to act as nature does as best you can. You are a surogate mother earth baby. Get used to it. I would suggest that you build yourself an incubator (Cooler, fish tank heater from wally world, large glass jar that the fish tank heater goes in, thermometer.) and let that keep your jars dark, and warm just like they like it. Have fun.

Pinning in vitro? That only really works with the PF "Mathias Romero" strain because it's a mutant freak. :) What you're simulting in a spawn jar is the inside of a cow's gut. You keep it warm, dark, and moist without any airflow. Once you birth the cake, you're simulating what happens when the cow drops the cowpie. It's exposed to air and light, and is much cooler.

Give your jars as much-or as little- light as you want. Pinning invitro is hit-and-miss at best. Most strains will fully colonize the cake well before any 'invitro pinning'.

while max is correct about finishing colonization b4 invitro pinning occurs, it is simply not true that only pf's pin invitro, or that it's unreliable. every strain i've tested, 15 in all, has pinned invitro every time. after exposure to light approx. 3 weeks.

so let me get this right I want to put my jars in the dark until fully colonized at 70 - 80 degrees before pinning starts I take the cakes out and I want to case them in my terrarium where they are then exposed to light.

Correct. 80?F is better than 70?F. Read P. cubensis Growth Parameters .

Thanks Anno I just read on a another thread that it is hard to get Puertos to pin. That is the type of syringe I have. If this is true is there an easier strain of P cubensis to grow for a beginner.
