Agar vs LC clone?
    #9197780 -

Is there a reason to use agar over Lc to clone?  I have been using Lc because it is cheep and easy for me to get everything and it seems to work fine.

Agar has the advantage that if have contamination, you can transfer mycelium furthers from the contam to a new dish, repeat until you have a clean sample.

also, fruits from multipore growth can be made up of more than one substrain. On agar you can isolate .

Hi MrBFrog: We have been blessed for ten years or more in that there has been a very clean supply of spore prints from mushrooms that have been grow under near perfect conditions in the Netherlands. As the laws have been changing there over the sell of fresh shrooms the supply of clean spore prints is going to dry up over time. The spore print part of the business is just a small part of how they were making money and as the growers move into other areas over time. We have see this in this country as I know of no one from the 80's growing boom that's still in the the shroom trade, they all have moved on.

So it's my option that knowing how to do agar plating and plate to plate transfers once again is going to be a needed knowledge and a most useful skill.

Again This Is Just My Option; Hipster

The advantage with agar is that you can see if there is a contaminant there. An LC can look clean and still have a contam that doesn't show up until after you inoculate something with it.

A:rolleyes:

i forgot to add that part, i was thinking of that half way through typing my post.

What does a contaminated LC look like? like a healthy LC. The only other indicaiton of contamination is a cloudy mixture. which usually means bacteria.

Not only that but the piece of flesh that you put on the LC or agar probably will have multiple substrains, some better then others. In the LC, you will have a slurry of multiple substrains so you might suck up a shitty one. On the other hand, with the agar, you can see the more vigorous and rhizomorphic substrains and then do sectoring to isolate the best ones. You can then add a chunk of the final agar plate to LC and have consistent results. This plate and LC can be turned into more plates and LCs and give you a virtually unlimited amount of culture to use.

I once cloned a monster by just throwing flesh into liquid and the LC didn't give consistent results under the same conditions. Now I use the grocery store agar tek to isolate monocultures then throw those into LC.
