Yes, peptone is worth the extra money.
The mycelium grows much faster on peptone agar

What about adding Peptone to your substrate if agar is not used?

Drip, Peptone can be gotten from www.fungi.com for $18 (100G).

peptone or ammonino acid is to supply nitrogen to the medium.
some fungus can use the ammonium ion, and some may use nitrates. i'm not going into detail but some need it and some don't. Nutritional requirements depends on its habit, grows on wood, dung, buried wood and so on.

If using the Rye Grain recipe in TMC, how much peptone should be added / quart jar?

How much should be used / liter of agar?
