What kind of peptone to use???

Hello Everybody!

So I'm looking through a telephone book thick science/lab/chem suppliers catalog and I'm thinking to myself, "Lana, what kind of peptone is best for mycelial growth?"

Bacteriological Peptone, Casein Peptone, Dipeptone, Gelatine Peptone, Meat Peptone, Proteose Peptone, Soya Peptone etc.

What tough choices to make!

So I'm throwing this out there...

What kind of peptone do you use, and/or which is the best to use?

I would go with the soy peptone, but that's just me. Any of the above should be a good source of protein, though, except maybe the gelatin peptone -- I have no clue what that is.

Peptone is just a peptic digest of something, and since fungi like soy this could be useful. It would probably be best to use it with a media that's not soy based.

Yes - soya peptone is used in agar for mushroom cultures.
I just ordered some to see if it really helps.

Zeronio, keep me posted on what you think.  I've read that most ... well, here's a segament of what I've been reading, if you're interested in the rest just ask.  I had to turn this into an image becuase I don't know how to convert a pdf file that has tables into text with tables :smile:

It said in that article somewhere that P. sinclairii mycelium grows best at a C:N ratio of 1:1 -- I don't know if that can be applied to most fungi -- I thought it was usually somewhere around 10:1. It also prefers sucrose media to starch which seems a bit odd. It's saprophytic, but also also an enthomopathogenic fungus (attacks insects -- I think this one in particular attacks larvae) so it's probably a bit different as far as nutrition is concerned vs. something that's only a saprophyte....

I also have a conflicting article about the growth rates -- PubMed something about exopolysaccharide production in Paecilomyces sinclairii -- their results are different. If you want the article I can email it to you as an attachment on Monday.


I think the best thing to do would be trial and error. Like I said above, peptone is just a source of protein, so any peptone should work. The efficiency of each type would probably depend on the specific fungal requirements for different types of proteins at whatever amounts and the type of media you're using and what it might be lacking. It can still break the protein down, though. Just try to keep the C:N ratio around 10:1.

The thing that's worked best for me as an additive, though, is a non-essential amino acid mix (I used the one for D-MEM @ 1/2 X) -- worked wonders -- beautiful rhizomorphic mycelium.

The one Stamets sells is from SIGMA

Sorry -- forgot about this thread. For some reason pictures aren't coming up, but in the table the C/N ratio for the highest dry mass was 20:1

This is a good table to have for reference.

I know this is a pretty old thread but I might be willing to revive this.

I am performing some tests with and without soy peptone.  Nitrogen and pH levels look moderate for soy pept.

I'll have some results in a few weeks.  By then I may have found a better thread to link to.

What kinds of effect does introducing peptone have? At what stage do you introduce it? Is this your first time using peptones of any kind?

My understanding is that peptones are most often used in agar or liquid media recipes.

I don't think it makes much sense to use for standard hobbyist mushroom cultivation (since it's expensive and helps other bacteria and fungi as much as the mushrooms you want to grow if your sterile technique isn't perfect), but it's a common ingredient in media recipes for various kinds of microbiology work.

I would look at maybe learning to use some antibiotics instead of peptone. Peptone makes everything spread faster even contamination. Fish antibiotics work great for the same purpose. I use two antibiotics Aqua-Mox (Amoxicillin) and Aqua-Ceph (Cephalexin), add a couple drops of McCormick's blue food coloring to your favorite agar recipe and you've got something that will help slow bacteria's and other contaminates and let the mycelium spread faster. Looks cool too. No more bare spots where nothing will grow on your plates because of bacteria. Anyways, check antibiotics out if you get time.

So far I tried using soy peptone for a malt - dextrose LCM.  Added with the dextrose and then autoclaved for 1.5 hours at 15 psi.

I did get significantly faster growth in the LCM.

I agree that it probably isn't necessary unless you need a quick turn around or there is a need for extra nitrogen.

I'll keep you posted on if I detect a bacterial infection and maybe have some pictures soon.
