Hi all,

I really hope I'm not posting a tired question here - I've been lurking mostly over the last 4-6 months and don't think I've seen this popped up - then again, this place is so god damn big! anyway...

Question:

if I have taken a couple of wild prints of ps. subaeruginosa (and sterilizing the card i printed on before hand), wouldn't the card/print be covered in millions of bacteria, rendering the spore print unuseable?  Or is it because there are a heap of mushroom spores on the print, which will overtake the other bacteria present?

I never tried that strain before, but everytime I tried using a print from wild cubes around here, it always got cobweb mold along with sometimes green mold. I gave up after about the 5th try and just cloned a wild cube, but that ended up getting cobweb too but I then cut out a section of the good growth and used that in some other jars.

matt - i did sterilise the card before printing... so that's good.

but - wouldn't putting the print in the oven @ 400 kill the spores off?

he ment cook it before the print. you'd more likely want to dip a peice of the inner stalk in an h2o2 solution and clone on agar.

I don't know why you would think it is covered in millions of bacteria. Where did you pick the mushooms, in the middle of a sludge pond? You did pick the cap of a nice healthy specimen and handled it carefully right?

It's fun to attempt to grow from wild prints but using agar methods is your best bet.

Yes, did handle carefully, and since the print was taken off one of the only wild shrooms still growing in the area it was the healthiest I could find (but maybe a tad small). The reason I assumed it had shitloads of bacteria is because there is germs everywhere and I thought if my print hadn't been kept in a sterile baggy or something it would have shitloads of bacteria.

Subaeruginosa is my favourite as im working with about eight strains - ranging from Tasmaniana, Australiana, the wavy cap and now a few unknown varieties.
Because your prints are wild, id suggest taking them with as much sterility as possible, then streaking on agar. If they dont germinate without being covered in strange liquids (common with wild prints from all around the globe) you can add a drop of gentamycin sulphate to the agar mix.
Either way, as SOON as they germinate and before contams get a chance, transfer to strongly peroxidated agar 6-7ml per 250mls of agar even! This kills the contam spores, then its just a matter of repeating until you have a rhizomorphic strain/ and your sure that all contam spores are dead.
Then just use an agar wedge to innoculate a mix of soaked hardwoods and brf. Use this to spawn bulk amounts of pine, red gum and other eucalypta wood chips and make an outdoor bed.

went back over the fakyou and there it is in blacknwhite.. but alas - i had taped the sides of the card and stored in a book so it should still be cool.

have another question re subies if you're keen - my shroom patch has well and truely diminished (actually, it was more like a shroom place, scattered with shitloads of little patches, not one or two 200+ shroom patches) - but when they were growing I transfered some of the woodchips into a pot and finished growing the shrooms out to a nice size. I still have the pot n woodchips but its completely dry. Alot of the woods still seem colonized but are completely dry. Can I rehydrate the woodchips and inocculate jars pf style (and then transfer outdoors in due time)?

i've had it probably stone dry for 2 months now - i've noticed some of the mycelium has browned - and some (on the other side of the pot) is white... do you think 2 months warrants rehydration?

really appreciate your input mate - i'll give it a bash  :wink:

I am about to cruise out for a bit but will try to explain the different varieties/locations/environment/etc when i get back.  I still have quite a few dried specimens and (as you have helped to ascertain) a (probably) contaminated print I could send you.  I know you'll be interested to hear I find them in the Eastern suburbs of Vic and am sure i have picked at least three to four variations - and after searching for all sub related material on this board, I read some of your writings on the wavy capped variation, which I can confirm, both my bro, good mate and myself have consumed at various times with the same conclusion: a tad squirmy.

Anyway off I go - will post variations and descriptions soon - PM me if you're interested in the print/specimen.

just went to grab a can out of the freezer and noticed i'd put a couple in the freezer - the colour is a tad off and some blacking/rotting.  you're welcome to these as well, however offputting they may sound  :grin:
