Simple filtered air glove box construction with Pics

Warning! doing this STINKS!and will set off smoke alarms.
I had all the windows wide open for these indoor pics, I suggest you do this outside if you try it.
This is a neat little filtered air glove box with twist off/on arm hole covers.

Get a suitable coffee can, this is a maxwell house one with the label peeled off.Heat it up on the stove element and press it into the side of the glove box with pliers or whatever.Make it wide enough you can work comfortably, my first attempt was too close together so I turned it around and did the other side.


Get a couple of those Glad blue topped twist lid things and stick em in the holes, they're tapered so they should bottom out about 1/2 inch before the twist off part.Mark around them with a marker while they are stuck in the holes. The ones in the pic are a cheap knock off brand I got at the dollar store but they are the same shape.

Heat up a knife and cut the containers a little longer than the marker line.It will take a couple tries cause the knife cools down....just reheat and press down steady, don't saw side to side.

Mix up some 5 minute epoxy and glue them in or silicone them in place into the holes

You can wear or add tyvek sleeves with hose clamps if you like or just use long gloves
Still makes a nice storage tote when not in use!
All these containers are polypropylene,and are a bitch to cut.(I shattered my first box trying to cut it with a utility knife) Melting works fine though.Look for a triangle with a 5 or "PP" inside it on the bottom of the container.
I have some PP containers full of BRF and verm (they can be PCd) and I'm going to give them a try for growing cakes.
all the glad PP containers have a nice taper so it should be really easy to pop them out when they are done.

The screw lid Tupperware holes are great. The ingenuity of the fellers here is so cool ...

Coffee can heated cutter also Nice.

ps ... could do another hole and Tupperware screw lid on the opposite side or end and cover with tyvek (hose clamp) for clean air supply to evaporate alcohol, oust, etc.

I can't take credit for the coffee can thing, stumbled on it in here somewhere.
I'm going to glue a couple more in the back with double tyvec filters screwed on...possibly one with a small hole in it so I can jab the scalpel/needle out of the box and put it in a flame to sterilize.
I haven't worked out the details yet.

These are the glad PP (polypropylene) containers with the BRF cakes in em.
I think they should colonize a little faster than round jars cause they got a thinner cross section.
I plan on shooting each end and each side with LC.
Hot flame sterilized needle right thru the plastic lid and tape after to shut the holes.

Here is the positive pressure filter system.

The box and pump...I need a T or second pump for the second filter...dunno yet though.

I covered the holes in the handles with micropore tape so the air leaving around the seam of the lid has more speed.

This is the high volume filter...One layer of Tyvek stuck between two rings. screwed onto (and epoxied to) a jar with the hose epoxied into a tight fitting hole in the tyvek.Glue one side of the hole in the tyvek with a fillet let it set then do the other side with a fillet of epoxy, after the glue sets scuff up the rim of the jar with sandpaper or a diamond grit sharpening stone glue on a ring and let it set..when that sets, glue the disk stuck to the hose between that and another upside down ring.

This is the high filtration system, I'll leave it on just to keep the box very clean when not in use but the flow is too slow to work with..
It's a small container filled with poly fill (which probably does nothing) and two disks of Tyvek epoxied into the lid...about a one inch hole is in the lid to let the filtered air out.The tubing is epoxied into a tight fitting hole in the base as you can see.The flow was very slow so I made the jar one too cause I wanted a little breeze.

this is the Exit. I'll use it to dump stuff out of the box or sometimes put stuff in it while I am working in it. Saves me having to lift the lid but if I need something I'll have to yell for it.
I may also keep the Bunsen burner close to it so I can poke the knife out to sterilize instead of drawing my arm out.

I haven't figured out what to do with the other hole yet...it'll probably stay shut.
I've yet to add the permanent gloves more pics later.
To use the box I'd load it with the stuff I need wipe it all down, spray it with alcohol/bleach spray the filters with alcohol, let it sit for a while with the filters blowing to get the fumes out then use it.
when not in use I'll leave the high filter running just to keep it clean.

I'm gonna set it up and leave an uncovered agar dish in it for a day then see if anything grows to test it. If it fails the naked agar test I'll put the pump(s) in a filter box so it draws air thru a large tyvek filter in the lid of the box.

More pics later when I track down long gloves or Tyvek sleeves.

I hope you enjoy the post, I think it will go a long way towards advancing my sterile techniques and cutting back on the amount of room I need to do sterile work.Previously I filtered an entire room, lately I just work quick and dirty, I think this is a good compromise.
And as pictured it is about $30 of materials.

Hey, I decided after taking a look at this to make one of my own.

I got an 8 dollar PP container and burned two holes into it with a pasta sauce container. I was originally planning on using the same sort of plastic drink containers but I didn't quite get the right ones so I opted for a straight gloves-attached-to-holes approach.

What I'm wondering is:
a) Do I need airflow when innoculating the jars?
b) Could I continue to use this glove box as an incubation chamber as well or would that require me to implement some sort of airflow?

Gloveboxes should be still air and NEVER have a fan or pump attached. Doing so will cause turbulence and INCREASE your rate of contamination.

as the above user said, your gb should be still as possible. gb prevents air which has contams to enter. it provides you with a sterile environment to work.

That's what I assumed as I've noticed a lot of the same replies around the forum. So in that case, would it be safe to assume that it would not be a suitable incubator as there should be gas exchange in an incubator?

Well I'd have to call my box an experiment. stick with still air.If I get good or bad results I'll post them, decide to add a pump after that if you still want one.
I guess you could use it to incubate (but I assume you ment fruting chamber)..just take out the gloves and add wads of poly fill for gas exchange.
But you'd be without a glove box for the duration and if your grow gets contaminated you got that in your glove box too.
Moving air (even filtered air) goes against the grain here, but I like to try different stuff.
If I wanted to paint by numbers I'd do something else.
