What's the fastest way to colonize a jar?

Just wondering what would be the fastest method for colonizing a jar? I'm guessing it would be rye mixed with coffee grounds and inoculation using an LC.

Am I right?

Ive never mixed coffee in my rye but I have heard it works to speed things up,I would say that the best thing you can do to help things along it to use a fast colonizing strain to began with and keep it at optimal temperatures

Yep, from my limited experience temps are very crucial. I have no Incubator and seeing as its winter here we get huge fluctuations in temp (40-80). I see huge growth spurts in my WBS colonization when it warms up.

I waited like 6 days for my spores to germinate so LC's ftw...

As for a fast colonizer you want a strain thats very rhizomorphic right?

The reason I'm asking this is because I have just attempted my second grow and found that another two of my 6 PF jars have contaminated. I'm pretty sure it's because of my temptation to open my closet to look at them each day exposing the holes to open air that did it rather than my sterlile procedures.

I can deal with failure, what makes it so faustrating is the fact that I have been waiting for a month for them to colonize, and my two best jars contaminated now when they were 85-90% done. This legnth of time makes me think that it's my own curiosity in checking them everydy that led to them contaminating.

...Anyway back to the point I wanted to find out the quickest possible to to colonize jars in future to reduce the risk of them contaminating, plus waiting for PF jars to colonize is so faustrating!

oh, and the strain I've got is B+. I know it's not a very fast colonizer but I may as well make an LC out of the 1.5cc I've got left in my syringe before I spend £15 on another...

An LC is an absolute MUST. I have 2 LC's hanging out right now in the incubator and about to move to the fridge.

As for the speedy colonization. Its all temperatures. Keep them optimal and the growth will most def reflect it. I keep mine at a constant 82-84. My BRF jars were completed in about 2 weeks flat and given about another week to just sit and fully colonize the inside.

Just keep the moisture content correct when making them in the first place and the temperture at optimal. Oh yea, and when using an LC to inoculate I personlly like to cut back just a tad on the original amount of water I put in the jar when I first mix it all up... seems to balance out the excessive water introduced with the LC.

I am no expert but I don't know about this.... Do you have aluminum foil over your jars? I look at my jars everyday and have never contaminated them this way. I suspect that if you left them uncovered this was possible. Your jars should not take that long to colonize. My times are 2 weeks for surface to be covered and extra 5 days for the inside. I keep my jars between 82-84. I find this ideal.

Since you haven't had PF success try grain. I am on my first grain and already going much better. Plus you can shake grain up to spread mycelium more evenly. Don't listen to anyone that says whole grains are harder. They are just as easy as PF to make. Plus you get way more subtrate to work with.

I like to follow the more experienced hands on this site rather than do experiementing on my own. I will get there but I just starting as well. If you haven't already read the link in Blutjager's signature. Also, CreamCorn has a great LC tek.

LC inoculated into a wbs/grain quart jar.
Wbs/grain moisture level must be optimal.
(a tad dry)
About 3 ml of stir plate mixed LC.
(creates finer myc particles = more contact points)
Once inoculated, gently & slowly roll jar, 1/4 turn a time.
(or whatever suits you)
Object is to spread any excess moisture into the wbs/grains.
Shake jar ONCE @ 20% colonization
Fully colonized in 7 to 10 days. :thumbup:

LC on WBS took a week, Spores on WBS took two and a half weeks. LC rocks and is crazy easy
Why would you need to sanatize the air and wash your hands if your looking at a jar thats most likely got a tyvek/poly and coffee filter on it? I love to sneak a peak every few days

stop peekin, lol get you some lids cover with tyvek and apply ring with a RTV gasket sealed hole about 1/5.. thats how i do mine, i peek atleast 3 times a day to see temps, because i too also have adjust temp settings throughout this winter i noticed since your using a closet getting a bigger bulb for your closet light, i was using a 120 watt bulb that kept my closest at 70 degrees but if it got cold i was fucked, so i just put a 200 watt bulb in and now it gets 85 degrees, so if its cold i turn the light on, if its hot i turn it off, using my noggin... i envy you LC users! you bastards! hahaha im stilling using spore syringes! which TAKES AWFULLY LONG TIME but not as long ass the shit that comes to me from TENN! hahah also befor i go into my closest i take a shower in ANTI BAC soap... NEVER BE too SAFE also spray the room with oust...... i heard carpets a good contributer to contams use some plastic on the floor wear clean clothes and make sure you heads washed.. and AGAR!! does that rolling a fourth of a turn work for spore syringes?

well a friend of mine inoculated straight pf style 17 days ago and had it incubating with a lizard heating pad in a huge 60 quart tub. it seemed to stay warm in there and he was excited to see his first peep of myco porn, softcore style. He recently made a TiT with the high dollar 20 gallon water heater. he keeps the water nearly 89 degrees. He filled his TiT with newly inoculated jars substituting 1/4 of the water in each cake for brewed coffee. His new batch is 10 days old and has certainly passed the older batch with nearly 100% colonized on the outside. The coffee and TiT has made a marked difference. Also He loves the Germ-X hand sanitizer that wipes into your hands. He uses it whenever he peeks at the babies.

In fact, fresh air is required for the mycelium to grow. A substrate that is too wet takes a long time to colonize, as will a substrate without proper gas exchange, both of which favor contaminants.

Looking at your jars does not cause them to get contaminated because they have a filter on them. Air in the incubating area doesn't contaminate jars for the same reason. In fact, fresh air is required for the mycelium to grow. A substrate that is too wet takes a long time to colonize, as will a substrate without proper gas exchange, both of which favor contaminants.

Now, I've been leaving my jars 100% air-tight sealed, and I've never heard any divergent advice in regards to this. Now, it very much makes sense that fresh air is necessary, but I assumed an "airy" substrate would suffice, which I always aim for.

My current jars all have the four pinholes in their lids, and a 1/4" to 1/2" layer of fine vermiculite as a filter. Since the filter -is- there, would you advise removing the seals and allowing some gas exchange, or will an airy substrate suffice after all? If so, I'm assuming I should at least wait until the jars are at least a good bit colonized, to reduce the chance of contamination, is that a correct assumption? Thanks much for any light you can shed on this.

RR says (and this is in very general non scientific terms) not to shake grains after inoculation so spores can germinate and merge which results in hyphae that grow faster since they are merged (as opposed to 1 spore with 1 strand coming out searching for a 'mate'). Once the spores germinate and the hyphae 'mates' you can shake and it'll speedup faster than if you shook after inoculating
