Im going to grow sclerotia invitro. ideas?

Ok, so i've decided to investigate the virtues of Ps. mexicana and invitro sclerotia formation. These are my plans:
5 one-pint jars.
2 CC spore solution (A-strain) per jar. (should i use more juice?)
Substrate will be: 1 part rye grass seed to 3 parts water.
pressure cooked for 30-45 mins.
Micron filter discs on the jars to allow for air circulation.
After colonization and trashing of contaminated jars, they will be placed in a dark cabinet and allowed to chill there for a couple months, blowing some air into the cabinet every once in a while to insure fresh air.
when it seems that sclerotia is done forming, i will harvest the jars.
does this seem good to everyone? are there any suggestions you people have for me and my experiment?

Use larger jars if possible, leave 1/4 or more of the jar as empty air space (keep in mind that the seed swells when hydrated during pressure cooking).
Ratio of seed to water is 2:1 not 1:3 (unless you are doing a steeping and draining method)
Pressure cook for a full hour at 15 PSI
Filter disk is fine but these jars will be incubating a long time and will dry out if the hole the filter disk is covering is too large. Try a 1 centimeter hole or smaller and cap the top of the jar with foil.
Blowing air in isn't helpful
Other than that your method is perfect.

Innoculate sterilized ryeseed. Wait three months and harvest. That is the beauty of invitro sclerotia production. Wasn't trying to be a dick.
Larger jars are better, larger yields per jar. Filters are not absolutely neccessary, but alittle clean gas exchange can't hurt.
If you really want to get crazy, Use two of your healthiest looking pint jars to innoculate 14 more pint jars.
Sclerotia are best consumed fresh.
Expect yields of 15-30 dry grams per cup of dry ryeseed. Very High Yielding.
