Melatonin as key to supercharge potency Report.
    #4886735 -

I caught onto the previous thread about how melatonin and the active ingrident in shrooms have a very similar chemical composition, thus supercharging potency , in theory. well anyways this is what i did. i took some cakes already colonized and cased them with coir that had a handful of melatonin crushed and added into it. i did many casing and lost track of what a i did to many of them. i passed on some of my casing to my friends. while the casings at his house what growing, i tried some of the mushrooms that ive been saving up(it was maybe 10 or more good sized shrooms). it wasnt great, not as what i expected. My technices for potency needed work. So, weeks later going to work and back i stopped by my friends to see the other casing there. turns out he did them with some other friends before i could grab them and save them. Fucking Stoner. i said to him, hey! your wasting them. its not enough for a trip. I did more the other day and it didn't do much. he said that he defently got a body high and that he saw the 'devil's eyes' in a cd case. and the two other friends had a decent trip as well. i thought to myself, how can this be when i did 10 shrooms to myself and he did 7 (estimated) shrooms between 3 people. Then it hit me. i may have put melatonin into those casing at his house. its the only explination i can come up with. Sense this i have been putting a shit load of melatonin into everything i make. currently have Pans growing on Cow Manure with melatonin. Put a shit load until there is a fine white tint in the manure. hopefully i will have some monster supercharged pan mushrooms. will let you guys know how it goes.

That's interesting. But for future reference, split your post into paragraphs, makes it easier to read.

And also, giving casings to friends? Sure that's a good idea?

I wouldn't say that the melatonin would be the only explanation. Shrooms vary from flush to flush and mushroom to mushroom. You can eat the same amount of the same strain from the same casing and get 2 different tripping experiences. Alot of it can have to do with the setting and your mood too.

Tolerance also. I think that tolerance to psiloybin relies mainly on the levels of serotonin people have in their systems. I've been looking into Monoamine Oxidase Inhibitors, MAOI, and they accentuate the potency of a trip upto 3 fold, because they build up a hell of alot of serotonin and slow the processing of psilocybin. In industry MAOIs are used to treat people with depression and sleep problems and come in pill form, where they require higher levels of dopamin, serotonin and other shit to make them feel better. Serotonin is in fact 5HT - 5HydroxyTryptamine. I know that by adding serotonin and melatonin inducing foods primarily before a trip, it will be much stronger, yet the scientific validity of adding tryptophans or whatever directly into a substrate isn't exactly established. If anyone is planning to use MAOIs, BE CAREFUL, READ UP FIRST!!! I don't want anyone dying or becoming schizophrenic and pointing the finger at me :-P

Interesting, I wouldn't be surprised if the enzymes for psilocybin production in the mushroom metabolise melatonin, which is 5-MeO-n-acetyl tryptamine.

Likely as not, the resulting psilocin analogue produced will be 5-MeO-n-acetyl-n-methyltryptamine, I don't believe it has been bioassayed as a single compound before, although I would be interested to see the subjective results of a few trials.

With 5-hydroxytryptophan, the conversion to psilocin is rate-limited by the amount of enzymes in the fungus to take tryptophan to tryptamine, and also by the amount of enzyme to methylate the resulting 4-HO-T.

I reckon that adding lots of melatonin would produce some interesting results, as well as the fact that it may well allow more total tryptamine to be produced, due to the fact that only one methyl group needs to be donated, as melatonin has an n-acetyl group on the terminal nitrogen.

Okay, if anyone is still here... I'm no chemist, but I've read that mushrooms will 4-hydroxylate any tryptamine, not tryptophan, as you mentioned.  Since MELATONIN is n-Acetyl-5-Methoxytryptamine, I thinks we should get 4-OH-5-MeO-Tryptamine?  Correct me if I'm wrong, but I sure hope that's what it is, cuz it has a nice ring to it.  (again, not a chemist, but is that a pun?)

HOWEVER, it is important to note:  Most melatonin pills contain only 300mcg melatonin.  [Would, say 3mg (10 pills worth) become a decent amount of psilocybin, or would this require growing in like pure pill powder?]
BTW, will magnesium stearate or croscarmellose affect medium?  Other ingredients seem to be plant derived cellulose and a sugar (dextrose).

Input?

wishful thinking, most likely. in the human metabolism, melatonin represents the second to last product of tryptophan metabolism via serotonin pathway (there are actually two end products). manipulation of melatonin into something active isn't really feasible.
and since enzymes are rather selective, there's no guaranteeing  melatonin would even be metabolized in mycelium

It's Alive! Honestly though why not just take melatonin orally instead of putting it in a substrate?

Lol too many reasons, so many variables..

Mushrooms are such sacred beings, some dare to say celestial, so i treat them how i would want to be treated, and in return they give me their love also.

Im gonna try it on a extra special batch of some cubensis i printed a while back

God that's hard to read.
Anyway 10 shrooms means nothing. 7 means nothing. Some are hollow, some not. Some are dense, some not ... etc.

Also potency can easily vary shroom to shroom and especially tub to tub.

You have to repeat the experiment multiple times using some very narrowed down substrains to even have a chance of this being worth time.
Anything else can easily be from hope or placebo. It's just not enough to say anything.

People have tried this with DMT with no results.
