Unsuccessful substrate innoc question

I am posting this question here, since I do not have perms to post in the substrate subsection of the cultivation forum. (don't post much, but it seems that would be the appropriate place for this, not sure how forum permissions work here.. but if anyone could explain this, I'd sure appreciate it.)

and the question:

I've successfully cultivated before (in a country where this is legal of course), but haven't made any attempts at this for a few years, until recently..

which is turning out to be my first unsuccessful attempt ever.

(which all events below would ofcourse be fictional events that my dog attempted if successful anyway)

I admit, I have taken some liberties with trying out a new substrate formula, and believe something here has foiled my attempt this time 'round.

I'd like to ask anyone's opinion of what of these ingredients, might have did me in on my inoculation attempt.

Today is day 10 since my initial innoc, and still no sign of growth, I'm quickly loosing hope that this will be successful.

I've worked with finch seed before, and signs of colonization have always happend by day 3 for me.

syringes were prepared from reputable vendor, of huautla pc species.

2 dozen 1/2 pint jars consisting of the following (approx) recipe:
(came up short on last 2 jars, so I eyeballed needed ratios)

8 cups finch seed
12 cups vermiculite
7 cups water
::::new items to my formula::::
2 grams L-tryptophan (attempt to jack up potency)
2 grams ground vitamin B-6 (needed to metabolize L-trypto, from what I've read)
1/2 cup fresh ground coffee (no additives, according to lable)


no pre-soak, but pc'd at 15psi for 45 minutes (batches of 6)
foil, with top layer of verm for contamination prevention

removed from pc and allowed to cool 4 hours b4 innoc attempt.

sterile work room, w/hepa filter, gloves, mask, lysol the works...

medium temp under jars are 86 degrees f.

I shared needle between jars, but wiped w/ lysol doused tissue between  injections, no signs of contam, anywhere.. so I'm suspicious of other ingredients maybe throwing off ph. or water content not being suffienct.

I left out calcium carbonate, because I hadn't recieved it in time for the mix (a day late from shipping), I'm guessing this may be what killed my batch, but my guesstimate was that most of these ingredients wouldn't have thrown it off that much.

any ideas would be extremely appreciated so I don't repeat my mistake here.

Do the shippers irradiate the mail now, that may kill pre-packaged syringes or prints? I know the last few years have changed shipment security, and if they try to kill dangerous spores of things used by bad guys, can we not then assume that enlightening spores of harmless things would be killed as well?

I put alot of effort and money into my attempt here and would like to learn from my failure.

Did you check the pH? And why coffee when you already got tryptophan?

If so, that's great.. These additional ingredients are new for me, where I've read of success elsewhere for others... I do not currently have a way to test the ph.

I guess I'll be investing in something that does that very soon.

I have no idea if coffee has trypt i was just curious why u ad it to the substrate.

I think a roll of pH indicating paper should be enough for a while..

Yup, I ordered it..


seems like a hugh oversight for the time and materials invested in a batch, when only like 95 cents for 100 strips...

I'd be willing to bet the B-6 vitamin through the ph off wildly.. I tasted one of the pills, and it's almost like a vitamin c tablet, with the acidity in the taste...

not very scientific, but this stuff adds up in cost quick.

(already at like $400-500 in expenses for my little project).

the coffee was a crazy notion I got from other posts regarding the caffine stimulating growth.
