You add it whenever you remove some. So like today I'm gonna go knock up 40 jars with 120cc's from two different cultures (60 from each) so I made 120 ml of karo (actually 200 at a time cause its easiest) and sterilized it then when I'm going to knock them up in a few hours I'll take 2 60cc syringes and suck up 60 cc's from the new karo I made today and inject 60ccs into each culture then suck up 60cc's. Then back in the incubator for a few days to consume the new karo then back in the fridge to store if it isn't used again by then.
