Testing an LC onto agar, should I do a drop then use a cell spreader to get more surface area?

Or do I want the drop to have to just grow out?

Either it needs some more time (Ive had this happen) or it was bacterial and the 2nd shake has spread the bact (Ive also had this happen)

No, I see growth but after the second shake the Mycelium looks weirdly but time will tell..

I'd like to add to this question. I've inoculated few plates with MSS, some were streaked with loop and some just drops left there. Ones streaked aren't showing any growth yet, while some with only drops started but very slowly. And one with drops, I see black spores on agar but nothing growing. Can it be that agar is a bit dry and that slows it?
I used no-pour, made ~15days ago.
MSS about a month old.
They've been in 68-76 temp.

I found my questions answer in the meantime
:muppet:
Im gonna squeeze the side of the LC barrel for a drop -as puff suggested to someone- then spread it  a little.

As for yours:

So you MADE the no pours 15 days ago and nocced the plates more recently?

I wouldn't think 15 days drying a plate that bad.

Had trouble germinating on hard agar. Soft, water agar is supreme.

You didn't add any info on the actual timeline but spores usually take a bit longer then when using clone tissue or agar transfers.

If spores were suspended in water (I presume vendor spores) they shouldn't really need a soft agar recipe. It's different with spore prints or swabs though.

15 day old agar plates is nothing out of the ordinary and 1 month old spores is also completely fine. Spores last a long time, 1 month is new spores.

Were you the one who made the syringes? Do you still have a print left? Where were you storing your MSS?

I would try 3 things:

1. You can drop a little liquid onto a swab and streak it on the agar β€” sometimes that helps a lot (or directly from the print).

2. For cases like this, I personally would opt for Pristine BRF Puck, because I’ve already had good results with two varieties in a similar situation. From there, you can transfer it to agar.

3. If you have a way to vortex the syringe, I'd do that if it's been stored for a month, just to homogenize the inoculum. You can make one out of a small motor (I'm not sure if a massage gun works for this).

Agar shouldn't dry out in 15 days.

Hugs

Thanks y'all for help! Sorry for not providing enough info. Streaked plates are a week old and droplet ones are 3 days old.

MSS is from the vendor, stored in dark and coolish place.

I did couple PF tek jars for the cake wars 6 days ago. Can't really tell if there's something growing on them, but one of syringes I sent directly to an extra spawn jar I had around and that one's showing spots of myc.
Edit: PF jars were noced in the middle.

I have made one syringe with sterilized water to try add few drops to those plates, so I might go ahead and do it. I'm aware spores take longer than LC/clone I just don't know whats the time frame.

Maybe I'm just impatient bastard seeking comfort, in which case I apologize for taking up your time.

I've had spores take as long as a month to germinate, and as little as 4 days. The ones that took longer were always cleaner.
And don't apologize for being impatient, the mushrooms will teach you patients. They always do.

Greetings friends, really want to get try running some pans moving into the next year.

I've attempted now twice before from LC to clean up on agar only to have jars stall.

Fingers crossed on the 3rd attempt.

I think my incubation temps were too low around 23c, this time will ensure a few degrees warmer.

Are lc syringes still worth trying if they are around a year old ? Or do I need a new culture?

Method: LC to agar , clean up transfers ideally 2, send to grain in smaller jars ...

Any tips would be super , mush love

I'd bet contam the problem more than temps. only way to know if lc is good is try. suggest just many more plates,.more cleaning,.posting more pics before inoculation

:whathesaid:

23Β°C will be fine for colonizing. Put a drop of LC on a plate by pressing the barrel and not the plunger. Leave the drop so you don't spread any contamination around the plate. If and when you see any sign of life you can place your plate with a slight tilt, if there is bacteria the mycelium will have a chance to crawl up the slope while bacteria fall in the other direction.

Baba just recently got his 3 year old LC to come back to life. Post pictures of the plate before sending to grains, pans are not as resilient against bacteria as cubes are so a clean plate is alpha omega.

I don't get it, did you buy a spore print or a spore vial/syringe?

You must forcefully hydrate spores if you bought a print.

3 days is nothing, but could start seeing something. A week I'd expect too se germination on properly hydrated spores ...

It's okay, thanks. Bigworm there got it resolved.

In my post I used abbreviation MSS, meaning multispore syringe.

Do you use agar at all? Or are you shooting spores into these?

First it was a swab to agar and it did not start.

Then it was a MSS from a trusted vendor that I shot a drop on agar, transferred to clean it up and took it to grain.  Consolidated and birthed, no dice, no fruit.

Most recent step was the MSS into a PF cake - took a piece of the myc to agar and am cleaning it up again.  The pf cake is sitting in a water tub, but not fruiting.

It’s very frustrating.  I have had plenty of rather dirty pf cakes fruit just fine.
