Will PC sterile water in 2 smaller mason jars then pouring them into a bigger one cause any issues?

I need some sterile water for some older spores I am trying to revive. I want to get a syringe and suck up some sterile water then suck up some spores in them.
My question is, I got a smaller pressure cooker and I want to have enough sterile water for a long time without using multiple jars. Could I PC two smaller jars then dump all the sterile water into a bigger one without contaminating the water? I highly doubt it but if anyone has done this with success I'd like to know. Thanks all :smile:

Is your bigger jar going to be sterile? Every time you open your jars the possibility of contamination appears. I can't tell you whether it will remain sterile or not since a lot depends on sterile technique. When trying to avoid contamination it is better not to put all eggs in the same basket.

I haven't really had any idea on how to sterilize the other jar. I was just going to rub it down with alcohol and pour some in, let it all evaporate, wait an hour or so, then do the same with bleach.

I'd do all that in a SAB then once it's dry, I'd pour the mason jars sterile water in the bigger one. Of course all that would be done in the SAB.

Think that would all do the trick?

It will be sanitized, but not sterile.

Spores aren't really sterile either, but your sterile water will no longer be sterile if you pour it into that jar.

All that being said, I'm not sure you're doing the best method for reviving stubborn spores.

Is it a print or swab? How old are the spores? What happens if you swab it to agar? Why do you think you need so much water?
