Tyndallization, thoughts and opinion?

Good afternoon!

I am wondering about something I kept reading about.

It seems that around these forums, fractional sterilization is very not recommended, or at least that's the feeling I got when reading. Why is that?


As context, I live in a 3rd world country where PC basically don't exist, and the cheapest lab autoclave is about $500. Insta-pots are 100 to 200 for the cheapest, small ones as well, and don't specify max pressure either.

I will eventually buy one, but it'll have to wait at least till next year so that I can get some money.


So I've been trying this method, sterilizing some grain jars and leaving them to the side without inoculation, just to check if the get contaminated. So far I've done about 10 popcorn jars and not even one had any contam issues after >2 weeks. Did I just get lucky?


So, is this method disliked mostly because it's inconvenient, or is it way too prone to failure/insufficient results? Is there anything that absolutely shouldn't be done this way that makes a PC super necessary?


What else can you tell me about this method? How do you personally feel about it?

I can't tell you anything about tyndallization and this is just a digression. If you want mushrooms you can always try PF-TEK. Not optimal but it is a very forgiving method and you don't need a pressure cooker.

I think it's a solid approach if other options are out of reach!  2 Week without any contamination is a good confirmation that what you're doing is working.  You didn't really lay out your process - multiple heatings with cooling in between?

I love that it's "effective in practice", but not reliable.  Makes it sound like a bumblebee - It works, but it shouldn't :laugh2:

I've experimented with long term (18+ hours) steaming of grain spawn with good results.  No cooling cycle.  Same process I run my bulk substrate (gourmets) through.

I have already golden teacher with PF tek in the past, and PF-to-bulk as well with good results.

After a few years of a break (and running out of shrooms) I came back recently, and wanted to go a bit bigger this time, so I'm putting quite a lot of effort in Agar. In the meantime, while the plates get ready, I am practising how to deal with grain.

My exact procedure is:

-Boil some popcorn. I simmer for around 60min after water gets to boiling.
-Strain/drain really well. I spread them out for further drying on a mesh or on paper towels afterwards
-Put them in jars, 60-70% full. I use mostly small jars because that's what I have the most of, but I also have a couple of bigger jars
-The jars have modified lids (4 holes, around 3mm each, protected against rust and covered above and below with micropore tape)
-Cover the lids with tinfoil to stop water from getting in when boiling/steaming
-Put them in a large pot with a lid. I put about 3-4 cm of water in the pot and fit 5-6 jars at a time (~400g dry popcorn equivalent)
-Boil for 30 min- Take them out, let them cool
-Next day, boil for 30min. Take them out, let them cool
-Next day, boil for 1h. Take jars out, they are done.

For big jars I use 50% longer times.

Then I store them without removing the foil, and check them every few days for any changes or signs of contamination, mainly because they are practice runs (and also because my agar is taking longer than I expected, my spores were 3-4 years old and took almost 1 month to start growing on the plates)

I find the process quite convenient actually, since I cook on an electric stovetop with a timer.

I don't have PC options at the moment so I'll keep going my own way for now, but you have satisfied my curiosity about the process!
