So if my regular mini monotub normally uses 4 qt of substrate and 1-2 myco qt of spawn I could instead just mix the whole thing at once and sterilize everything in 6 jars then add my agar wedges and skip the grain spawn part.

So everything being sterilized from the start perhaps would increase success rate.

Then once they are colonized break them up and add to my monotub and case them right away?

Could be interesting to try I like the idea of having the whole thing sterilized and colonised right away. Maybe it will be a bit faster too since you wait less for grain spawn and then colonization in tubs.

I usually go heavy on the spawn, like 1:2 grain substrate, so you could even just do like two or three of those and then spawn to three qts of coir ammended with worm castings.

This is essentially how I plan to make outdoor beds this year. But the same concept works for a monotub.

Sterilize the manure or compost because it's more of a risk and makes grains more forgiving anyway.  Pasteurize worm castings because it's less of a contamination risk.

The worm castings will encourage the mycelium to colonize the coir well too.

But I meant I could try to mix everything together in jars like your aio technique including the worm casting to so it has to be sterilized with everything not pasteurized.

I dont see how I can do aio jars if the worm casting must be pasteurized and not sterilized I thought we just mix the coir, grains, manure and worm casting together in jars and pc them then add agar wedges with flowhood

Then break them once colonized and straight put them in the tub?

Either way can work.

But if you want to do multiple monos, you can do the manure and grain by themselves if you want and spawn it to substrate, rather than running six jars per tub.

That's all I was saying.  Both ways can work.

Oh ok I see. So you mean mix the grains with the manure and pc them. Is it ok to also just add the worm casting too in since its just a small amount might as well sterilize it with the rest.

Should the manure be on the dryer side of field capacity a bit when mixed with grains for the grain spawn.

Then bucket tek the coir only later.

Seems like a cool technique to minimize contam.

It depends on how dry your grains are.  If you don't want to dry your grains, then run your manure a bit dryer to compensate.

And sure you can sterilize the castings.  That's actually how I do it but I'm using smaller containers so I thought I would suggest a different approach in your case.

Yeah, I would like to experiment with it more. It really needs to be rinsed well and charged properly too.

In substrate it made cyans behave a little weird, sometimes not pinning and just wanting to make stones.  Other species were fine with it though.

It definitely improved the texture and aeration.  And should hold a lot of nutrients if I charge it right.

When I try it again I plan to run it in a thicker substrate.

Question of better Agar to transferes, cloning, put spores??

The recipe that I found more is:
500ml water, 10g agar , 10g LME..

But you say to grow better, you use only 2,5LME..

I make agars for first time in next week with LME..

If I use 5g LME to 500ml (1%)?  its a good compromise to cloning, put spores..?

I prefer to clone to 0% (just water and agar).  Spores germinate fine on 0.5%.  But 1% is fine for pans too.

You got some pro tips for long term pans storage in slants? What % nutrients etc.

I use cryovials with sterile distilled water when I store cultures long term and keep them at room temperature.

There's a thread here about it.  And it's one of the simplest and best storage methods generally, because sometimes tropical species don't like refrigeration (though this is more of an issue with some gourmets than copelandia).

I'm not sure but I run my mini fridge at 50f, and have pulled wedges off plates that are several months old as long as they don't dry out to much.
