The beginning of my Journey back into this hobby.. * 3

Picked these 3 up.
Gonna test them to make sure they are clean on agar, brf puck, and then transfer to LC for master LC's.

It's been a long minute since I've got back to this hobby so I also picked up some new tools to help me.

2x4 flowhood

Now I can get a chance to learn agar.

Induction 23qt presto & commercial cooker

Grateful for this gem.

So at the moment I'm gonna do a deep clean of my area. While working on cooking up some brf pucks, agar, and LCs.

As it's been a long while I'm sure I'll have questions and need to pick some of y'all's brains.

Goal is brf slurry to 5 racks of whole oats. Then 12 54qt tubs of coir. Or maybe I should do 1 of each tub so I don't have a complete freaking disaster. Ahh forget about it go big or go home. what y'all think? Either way I got a lot of work ahead of me before I get to that point.

"By 5 racks of whole oats I mean
1 rack = 1 case of jars
60 jars total"

Pucks, LCs, Agar



Better later than never.

Conditioned PC ring.

Turned induction on @ 1800w
Until steam was vented for 15 minutes then placed toggle on left it cooking @ 1800w until PC hit 17psi then dropped to 600w and turned timer on 60 mins for pucks

20 brf pucks
60 minutes @ 17 psi
3 cups Verm.
1.5 cups of fresh ground brown rice
1.5 cups of H20

3 LCs w/stir bars
30 minutes @ 17 psi
1/32 lme
1/64 peptone
1 tbsp light corn syrup


Blending dry ingredients in hot water before PCing.

Cooked set in cleanish tubs until ready to use.

Until steam was vented for 15 minutes then placed toggle on left it cooking @ 1800w until PC hit 17psi then dropped to 600w and turned timer on 25 mins for Agar.
Don't forget to boil your agar before you PC!

Mix dry ingredients in water and mix well. boil and turn off as soon as boil so no over boil. Then pour into media bottle and PC.

Agar
25 minutes @ 17 psi
500 ml of agar
7g lme
10g agar
500ml distilled water
20 4oz jars

In the cooker now. Got side tracked.


Some of the 4oz jars


Gonna cook new agar in media bottles and let cool then the next day I'm gonna heat up in my insta pot on sous vide for 30 mins at 180f cool to 120f and pour to new plates.


Or you can even go this route

So since I never ran agar before I was reading ReverendMyc's post on Primo plates and his recipe is:

That makes 0.5% nutes and 2% agar, I like it weak and hard, baby.


And from what I've read 500 ml makes 20 90mm plates. So think I'm gonna double this up so I can pour 40 plates total in the morning as I got 2 1000ml media bottles.

Also.

I'm gonna add this here to because I was wondering when to know the right temp is to pour and I just read the tad bit of knowledge.

When you can handle the agar bottle without burning your hand but it's not clumping , it's usually around 146 to 150 f , stack your plates and let them cool in front of your hood you may have a couple on top with condensation but it's no biggie set them aside for future use it will clear up

Bloody hell. I screwed up and fell asleep after my 60 min. cook. I suppose I should have set an alarm for a 90 min reminder.

Anyways I clicked the trusty search button above and searched can I recook my agar. I'm so glad it's not going to waste.

This is the answer I got.

60 minutes is way overkill for agar but yes you will be fine

So I'm struggling with this agar process.

I'm just gonna make prepours with the rest of these 20 4 oz jelly jars.


I don't have much more time to juggle with pour to these disposable plates.

This was my bad trying to go cheap with the 500 throw aways. Next time I purchase for the others reusable like mentioned in Tek.

Primo Plates: Reusable Clear Plastic Petri Plate Prepour tek +


Hell I might use these 4oz for a second though who knows. Anyways I'm wasting time writing off to drilling out the 4oz and placing stickers on so I can mix and cook.

Live and learn. Just grateful to be back at it again.

Noob mistake. Didn't follow the process.


Agar question?

Do I have to boil agar for it to solidify?

My natural hot water is hot as fuck so I assumed as long as I threw in a stir bar and let it run 5 minutes to dissolve everything I be ok. I poured to 4oz jars.

500ml water
10g agar
7g lme

I'm waiting for it to solidify then PC. It's been about 90 minutes.

So I finally took a step back.

Reassessed my thoughts. Realized I need to slow the fuck down. This is enjoy life not try to get to point A to point B in 2 seconds. Moreless I was causing myself more trouble trying to run before I was comfortable crawling.

Any ways I finally did it got my poured plates all off them clean.



I did give them a week while I was on vacation to make sure. Now time to do some transfers.


Picked up some new cultures also.

This one has some funk.

I'm gonna try and save it.

Then I got these.




So I got a lot of work but this time I'm not rushing everything.

Also I still have original stuff post up top which is gonna be my start as originally planned.

Nice work and clear write up. It's nice being able to follow along.

That first APE petri, with the two segments. What causes that to happen? I'll be getting into agar soon and I'm just wondering how that happens. You placed a transfer, it grew out, then there was another branching that happened from the original mycelium? or is it from different mycelium from the get go?

It seems to be that's the whispyness they talk of when looking for good agar transfers, so I would transfer a section of that whispy light mycelium on the left, from the top and bottom before it clashes more with the other. And then put those 2 transfers on 2 new plates. Those would be good master plates then right?

I'm kinda learning as I go.

From what I understand I want to take a transfer from both sectors and transfer to a new separate plate. Then grow both of those out and see which one I like best and keep that one.

Yea for sure transfer the original there's no shortage of sections to choose from, but I meant the whispy lighter one you're running out of real estate, so I would take a top and bottom transfer pronto from the light whispy one. I'm just not sure how it reflected that way in the first place or if that's common. Maybe a hyphael limb projected like an inchworm far and then it grew towards the original and created a boundary?

Still not sure what master plate means and if a transfer plate can still be a master plate, or if really slow nutrient composition is what makes a master plate so that the mycelium doesn't age as quickly.
