Fahts Easy pan cloning * 37
    #29263448 -

Yoyoyo party peeps.  So cloning pans can be daunting if you’re trying to get a tiny sample of inner tissue like you would larger species like cubes.  Here I’ll show you how to easily get a viable clone pretty much every time.

There’s things in this post that aren’t new at all like water agar plates etc but I haven’t seen anyone “skin” their samples and it works great.

1. First is getting a large tissue sample.  Because pans are so thin you can actually “skin” them.  This works best when they’re mature younger/close to mature (after doing many clones of pans this way, it may be easier to “break” the stipe in half when they’re older, but it’s better for sterility if they aren’t overly mature—you want white inner tissue.. if it’s yellow, it’s going to be very bacterial).  Take a scissors and cut like a 2-3” (or as long as you can) piece from the stipe.  You can use the very top with just the cap cut off but cut any fat base part off.  Ideally, you want it to be as straight as possible without any twists.. twists make later steps a bit more difficult.  Don’t worry about the scissors being super sterile.. you won’t be using tissue anywhere close to the ends.

2. I like to take a piece of paper towel that’s fairly wet with peroxide and gently run the stipe through it.  Here you can see the stipe is cleaner afterwards

3. Now take a scalpel and just start a split down the middle of one end.  The fatter end is better.  You can see the split started here

4. Now pull those splits apart so you have two halves.  Take one of those halves and bend both ends together so they’re touching.. bend them backwards so the inner tissue is out/up and the two outside “skin” ends touch. What will happen is the outside skin that is “bendy” will stay intact but the dense, stick-like inner tissue will snap.  You may have to help that snap along by crossing the two ends past each other.

5.  Now pull down on one of those sides/ends and you’ll effectively “skin” that inner tissue and you’ll end up with a huge chunk of that tissue sticking straight up.

6. Take a sterilized forceps and break off a chunk of that tissue.  It’s best to keep track of which side was facing the center of the stipe so you can put that side down on the plate.

Pan stipe “skin” isn’t very protective because of how thin it is and cracks do occur often when they’re mature (and even when not mature) so if you put this tissue to higher nute agar, you will still get bacteria along with myc coming off the tissue.  That’s why you want to use water agar or very low nute agar.

This inner tissue, while more than likely still bacterial, will rarely have mold spores on it.  I say rarely because I’m sure it’s possible but I haven’t run into it yet.

Bacteria is easily delt with more so than mold.. so this is why I use the inner “skinned” tissue rather than just placing a whole piece of stipe on agar.

For those that don’t know: Water agar is super easy.  You just make agar without nutes.  So just add your agar agar powder to dh2o and sterilize.  There is still a very minute amount of nutrition in agar agar powder so some initial bacteria can/does happen but because you use such a larger piece of tissue, the myc quickly overcomes any bacteria growth and it’s stunted near the tissue sample

So let the myc grow out a decent amount before taking any transfers.

Here you can see on this WA plate that there’s still some bacteria near the sample but stops quickly as the myc that grew through the agar came up through the surface as it grew out and hindered the bacteria from moving further.

You can also use very weak nute agar.  This is a plate that I used .7 of a gram for 300ml of dh2o.  The “L” stands for low nute. Edit:  I don’t even bother with cloning straight to low nute anymore.  I just use WA because it works so well

I’ll take any transfers from either style plate the water angar plate and immediately put it to a low nute cabin sequester.  This greatly reduces the amount of transfers you need to take.  If there is a small amount of bacteria in the transfer, 99.9999% of the time, one transfer put through a cabin will get you a clean culture.

I run everything through a cabin whether or not I think it’s clean… ms, clones, everything (all species) before I take it to grain.  That’s how much I like the method.  It has helped my final grows immensely and saves a hellava lot of time and plates.

Speaking of which, you’ll notice I put multiple clones to a single plate.  No reason to waste a single plate on one clone.

Hope this has helped some of ya!

EDIT 10/12/25: so it’s been a few months and thought I’d add some pics of the clones I’ve gotten with this method fruited out.  It works incredibly well.  Basically, if I clone to WA, I’m getting that clone.  Since I switched to cloning only to WA, I haven’t lost a single one.  I actually started by cloning 12 of each variety (bvi and peace river) thinking I was going to lose a good amount of them and I ended up with 22 clones total and was like, oh shit, I have to fruit these lol.  So now I only take 6 and probably will drop that down to 3 eventually.

Great little write up as usual Faht, I use almost the same method, but I use a scalpel only and haven't even thought about taking it to water plate first, I'll be sure to try that, thanks.

Here's the latest Pan clone I did.

Great write up!

Water agar to cabin is definitely the way.  And I'll give your peeling method a try again next time I clone.

I just send you are mssage asking what dope stuff you are currently up to. First thread I open after hitting send is you sharing this dope stuff:rockon:

Keep up the good work.

Very well written and thought out. Easy to understand.

I got some TTBVI clones currently going on water agar and brf pucks. Just to see if I can tell a difference and to experiment.

I will follow your advice and do a second transfer using a cabin:amusedapplause:

Faht you never cease to amaze me.  I have a hunch this would work with other thin stiped species as well, and I definitely plan on using this in the near future.  Many thanks man!

What are you using for cabin sequester? Coverslip or something else?

I just use an agar roof.  I’ve used cut slides themselves but they attracted a lot of condensation.  I’m thinking that the thinner coverslip does less of that.

Is this water agar for covering cabin or regular agar?

The water agar is just for cloning to but a WA roof doesn’t sound like a bad idea.  I just like cutting a roof from the same plate as the cabin

Oh, got it. Does it work good? I thought nutritious agar would be bad for covering cabin

It does work well.  There’s only been a couple out of many that I’ve done that I can’t seem to rid the culture of whatever is riding along.  I’m not even sure it’s bacteria tbh.  It doesn’t show up on the plate itself.. it’s more like it’s inside the culture because of the way the myc grows and 1 of the 2 I haven’t tried fruiting yet because I keep going back to agar instead of grain due to how it looks so that one could just be how the culture looks lol

Long story short, it works well enough for me due to ease but a coverslip or water agar roof will probably be the better route.  Try em all but you’ll probably find the nute roof to be sufficient.  If you wait long enough for the myc to get far enough away from the clone sample on the WA plate, you probably don’t even need the cabin.  I just figure since I’m doing a transfer away from the clone plate anyway, it might as well be to a cabin

Here’s some pics of the most recent clones.  Took 3 bbar clones and as of now, they look like they’re going to be successful.  The yellowing on the plate started showing up during the heat wave I had… showed up on most of my plates no matter if they were WA or nute plates and doesn’t seem to affect growth at all so I’m chalking it up to metabolites from stressed myc.  It appears to have gone with the heat as well.

The extra plug holes are because the first plug I took was too thick for the receiving plate and the plug would’ve sat above the walls of the cabin so, I had to move in closer to the center to get a thinner plug.  Usually when that happens, the plug will cause the roof to crack.  I’ve since fixed this problem by simply pushing the plug out of my punch a little bit and cutting half the plug off with a scalpel.  So no more broken roofs or plugs actually touching the roof.  It was a huge epiphany for me because that was very frustrating trying to match up donor plates with recurving plates that were thicker.. now I just cut the plug to size

Here’s the clone cultures I had problems with..  this ttbvi crapped out on me on the first plate run (it didn’t look well at all compared to the other clones).  I basically ran it through the cloning process again by putting a transfer to WA and then running it back through another CS.  That is the process I use for “fixing” any culture/the plates I’m about to show..

It’s still a crazy over-active culture so I’m apprehensive about running it but it does look much better than the first round.

This is the peace river clone that I ran and it crapped out and didn’t finish colonizing.

Again, it does look better than the first round but not much and there doesn’t appear to be visible bacteria.  These are .3% nute plates tho and it’s going pretty crazy so it’s probably going to be VERY crazy on high nute grains.  I’ve got 11 other clones that I took and aren’t acting like these cultures so I’m in the fence about even trying to run them and possibly wasting substrate and time.

This jambo culture is what’s got me even wondering if I should run the possibly “fixed” clones at all.  It’s ms and I’ve been trying to get it working for the last 2.5 months.  I’ve ran it 3 times now with the same result of crapping out during colonizing.. I’ve run it through the gambit of multiple CS’s and this last time I ran it through the cloning process..

Again, no visible bacteria and this does look a little better than previous runs but I’m going to let it grow across the trench and put that back through a CS before I put it to grain again.

Here’s an example of a culture I did “fix”.  It’s a bvi x Tex king.  This is how the culture looked when I got it and somewhere along the line it got fucked up.. it was growing thick and didnt look well on grain so I didn’t spawn it.  Put it back on WA and through a CS and it’s back to its original state
