Reviving dried agar and LM Mycelium
    #28741121 -

Two different topics, but both related. If it's better that I split them up, let me know.

1. When I did my first grow, I saved two chunks (from the stalk) out of my best mushrooms on agar (separate plates), put them in plastic bags, and stuck them in the fridge. One year later, the agar is dried out and I learned that wood splints should have been added to prevent the mycelium shutting down cellulose...eating. My brain forgets the right words.

Am I right in thinking these plates are done-for, or is there a reason to try and recover them? It was potato-starch agar, if it matters.

I searched on the forums and it seems like wood splints (basically a piece of wood in agar?) are the best way to store good clones long term. Or is there a better way? For reference, one monotub is enough mushrooms to last me like 2 years (improper drying ruined 1/2 my yield, but I know better now). I'm not doing anything crazy, just 4 quarts of grain into coco coir substrate. I guess I just don't use them heavily.

And that's why I'm asking. If I'm only going to be growing every 2 years, how do I keep my clones alive/viable with minimal maintenance? I'd really love to not have to make fresh agar every 3 months. How do I keep the agar from drying out? Should I just keep them in a quart jar so that it can be sealed in the fridge?

2. I know Staments is a mixed-bag on these forums, and his stack is thought to be legit to some, and nonsense to others. I'm a curious sort, but also, it appears that most people are using LM fruit body, rather than LM mycelium. Apparently the big benefits are in the mycelium rather than the fruit body for Lion Mane specifically. Am I right in thinking the best way to bulk-grow mycelium is to look into liquid culture methods? If I wanted to yield 2.5g of mycelium per week, would I need 1 quart jar of LC? 2? 20? Some idea of scale is all I'm really after.

This Mushroom Cultivation is for magic mushrooms, Gourmet and Medicinal Mushrooms is for, well, those. I think there is general consensus that LM is the latter, but one could start a philosophical discussion about that. There is also another forum space for that too haha. Anyways... Let admin decide about where to store. But there is some overlap regarding your mycelium question with this forum.

>>I think those are done for. But I'm not experienced enough to tell you for sure and you might be lucky (?).

>>There are other ways to store a clone. 'Better', is a matter of possibilities and also of taste. What I read is also that addition of splint would work. But I have no experience with that yet. For me something that takes less storage space would be better. Vials are mentioned a lot. Recently I learned about making mycelium in stasis or 'dormant' (not sure anymore actually whether that is technically a correct term...) by taking away oxygen and nutrients, at the moment the mycelium is alive. Like by putting the mycelium in water and sealing, in a small vial. But also it appears you could store dried grains.

>>I have the feeling it is also just very accidental and very convenient for supplement producers that instead of just only the fruiting bodies, also the mycelium is usable.

Anyways, what I understand is that there are more variables that make some contents (more) active. It is not just 'mycelium OR fruiting bodies', but more variables play a role. Just like the nutrients content label on a package for potatoes makes one conclude the nutritional value is always identical (regardless of time of the year, temp and sun conditions, soil quality, variety, NPK, etc.).

For actives for example (from magic mushrooms) it seems alkaloids seem to be more activated in mycelium when there is a threat, like mites. The same principle of influence of context conditions quite probably apply for lots of contents.

So, if you are talking about 'the best way'; do you mean BE or ABE? Like yield in grams or yields in grams of the specific chemicals you're looking for? Based on that, and practical possibilities, different answers about the best way.

I'm not familiar with extracting mycelium from LC. I think I've read some people do that for magic mushrooms. If there is a tek to extract mycelium from LC for magics and other mushrooms, I am keen to know. Distill the liquid and store the salt? Regarding bulk grow and yield, my guess is that for the same size jar of LC vs grain, grain jar will have more yield of mycelium. And additionally you have grains in your yield. One can then also state it has additional β-Glucans! Actually yesterday I dried LM substrate (with rye berries). For sure it is visibly less dense colonized then compared with cubes.

Thanks. I'll check those out.

...snip...

I genuinely have no idea what BE and ABE means. I checked the glossary, and asked google.

As for it being 'very accidental' that fact is not lost on me. There's a reason I'd rather grow my own than buy from him. He gets nothing if it doesn't work, and if it does, then I gain a little bit of faith in humanity.

When I talk about yield, I mean that the schedule for dosing is 5 days on, 3 days off. So within a 7 day period (because buffer) you need to be able to grow at least 2.5g of edible mycelium. That's the metric. I...see it no different than making cookies. You need X amount of butter. Either it grows that fast or it doesn't. And, sure, there's variability. Maybe my honey is different from yours. Maybe my water says it is distilled and it isn't. So maybe I end up needing to crank it up to get what I'm after. But if a quart jar grows so fast that I'd be looking at 50g a week from a quart jar, well, ok then.

Also, to clear up any confusion, you eat the fruiting bodies of the cubes, but the mycelium of the LM. And, yeah, I agree, convenient he happens to sell that. But I'm not buying from him, because I prefer to verify science when I can. I can buy lion's mane from the local farmer's market and get my myc going from there.

BE is Biological Efficiency and ABE is well I meant Actives Biological Efficiency. Like the chemicals you're looking for. But I might have made that abbrevation up. Not on purpose. Sorry.

Ok, so you're targeting for 2.5g dried LM per week. Clear. I think lots of capsules have just fruiting bodies + grain + whatever fillers. Anyways, 2.5 g that's 25 gram fruiting bodies (let's say conversion of wet to dry is 10). The grain mycelium I dried yesterday was 221g wet to 81g dry (so about 1:3). That was from a 600ml rye jar, of where I took only grains that were visibly  myceliated. It ran for 2.5 weeks. Anyways, further discussion now needs to go to the Gourmets section! J'accuse moi!

Regarding eating fruits from cubes, well, that's where I saw the overlap; also cube mycellium (not the fruit) has alkaloid actives. But now beware of shitting verm gold!

If the plates are still clean you can take a bit of the dried up agar and put it on a new plate.

I recently did this with a plate that was a couple years old and looked like a potato chip, came back to life in 2 days.

Cool. I'll give that a try. Just as an experiment, I left 6 plates out around my place. Most had their lids on (not airtight lids), but one didn't. Only one plate got contam on it before they dried completely, and that plate also had a little gnat-type creature that had crawled into it.
