This may be a dumb question... but how do you securely PC these tiny ass cryo tubes?

I usually have success standing them up inside a short half-pint jar.  I like to place a piece of  balled up paper towel or aluminum foil in there to help keep them in place, especially if you are not sterilizing enough vials to fill the jar with.  The piece of paper or foil compensates for the empty space.

My dude. Thank you. I'll use the lil jars from this tek just for that.

Thanks Mycoplex.

I have these creme brulee cups that are perfect for doing a bunch standing up at once. But any little jar will do.

Also doing new lids for the puck jars. 1/8 inch drill bit and cut up SDF shiny side up. Nothing startling. Just adding pictures to update the op when I get a round tuit.

Also tried using h2o2 to dip whole pieces of stipes from some pans before dropping them on the pucks. It slowed the recovery, but may have led to less bacterial clone transfers. I have transfers from dipped and un-dipped pucks on agar for analysis now. Will add the update to the op if it seems to "pan" out.

Undipped on left; h2o2 dipped on right.

Found a dumb thing I want to try  that could be a new use for pucks, if it works. Restarting a failed tub.

The only living culture I have of a strain I want to keep is in a tub that won't fruit. I have never gotten pan bisporus to fruit and used up the only swab I had long ago. No obvious contamination, just doesn't fruit. So I am going to take a few samples of the mycelium from a non-fruiting tub, put it to pucks -> agar plates -> muda jars -> top fruit.

Curious to find out if I just ended up with non-fruiting genetics or if it is possible that cleaning the myc up enough could result in fruit. The test won't prove anything with a negative result, but on the off chance that I do eventually end up with fruits it will be a way to save a grow gone bad.

I have actually tried taking spawned tub myc to agar plates in the past. It will come as no surprise that the resulting plates where disastrously atrocious. So it will be interesting to see if pucks hold up better. Maybe an h2o2 dip will be called for here as well.

Also taking some grain from some suspect grain jars to pucks before spawning them to my version of shoe boxes. Pants and suspenders kind of thing.

Time to get in the flow.

Fresh batch of pucks.

Suspect jars of grain: Pan Cyan BVI, Mexicana Chicon Nindo, Pan Cyan Brooks, Cubensis Golden Teacher (left to right).

Pan Bisporus spawned 44 days ago. Colonized and cased 28 days ago. I could just source some other spores and probably will if this fails. But where is the fun in that?

I guess I can triple post in my own thread.
So just some pictures from the excursion mentioned above.

First off, these are grain transfers from the suspect jars.

I hate myself a little for doing this, but I wanted to get the bell cap subtropicalis cloned asap, so I grabbed one well before it was mature. I set it in the flow, moved the tray out of the way, soaked my gloves in iso after handling the tub, tray and camera. Then chopped a couple of pieces of stipe  between the cap and bottom, dipped them in hydrogen peroxide and stuck them in puck jars. I used sterile procedures even though the samples were from a non-sterile source.

Then I did similar with the bisporus tub myc but instead of taking the sample from a mushroom, I grabbed a bit of myc from the corner of the tray.

The teal bowl has the h2o2 bath. In the last picture, the two on the right contain samples that were dipped in the h2o2 bath. The one on the left went straight to the puck with no bath.

Now we wait... again. Lots of waiting in this hobby. Time to make some gummies.

Some promising updates.

The transfer from bisporus tub substrate is progressing well for both of the h2o2 dipped and the undipped transfer samples.

Very hopeful about this trick.

The grain transfers from colonized jars are all taking off nicely.

These are GT, pan cyan brooks top and BVI bottom two center, & Mexicana Chicon Nindo from suspect colonized millet jars. Note that the grains are fuzzing up but not quite leaping off to the puck yet. I suspect this is because the grain still provides easily accessible nutrition for now.

And some cloned fruit transfers all dipped in h2o2 before dropping on the pucks doing their thing as well.

Subtropicalis Bell Cap and Pan Cambo Komodo. Note that the Komodo pucks were 10 weeks old when inoculated. I was worried that they might be too dry and even added a few milliliters of sterilized water to one of them. But both are taking off fine regardless of the added water. I also cleaned out several other expired jars prepared the same day and although the pucks had shrunk notably on some, all were still soft and moist to the touch. Both of these are leaping off of the transfer sample to the puck, which makes sense since the myc is looking for new sources of food.

From a 3 year old KSSS spore syringe, only 4 days after inoculation.

Pretty sure that black spot is a clump of spores, but let me know if I'm wrong.

I love these things! When nothing else will start a swab, try these. The container had a little extra water so I dipped the swab, streaked the plate, stuck it in the puck. Only one germinated.

Just like magic.

Since I wrote up the op I have become more and more convinced that these things solve or simplify so many steps in cult. Just so handy for germing, cloning and cleaning, why start anywhere else? I still use agar plates to confirm and inoculate, but this is where I start everything now. An extra step that can save so many.

^^Agreed, I was struggling to think of a simple way to bring back all my old spores and I stumbled upon this.
I just used two old Penis Envy swabs today, one on agar, and one I just shoved into brf puck.
I am not too confident about the swab jabbed into the puck but I guess we'll see how well it works if at all.

(my doubt is solely from not remembering when I took this swab and how clean it was)

Only one way to find out. This has been by far the most successful way I have found to germinate old swabs. Hope it works for you.

Here comes a whole lot of update pictures on the pucks that I have in progress. I will be doing transfers to agar plates from the leading edges of myc growth on each when I get back next week.

Pan Cambo Komodo

8 days from inoculation with a bits of stipe from a fruiting tub on pucks that were made 73 days earlier. So it seems safe to say that they keep for a while. You can see the blackened stipe sections in the pictures. I didn't try to take inner material from these tiny stems. I just dipped whole pieces in h2o2 then dropped them on one side of the puck with plans to pull some clean myc from the other side when it gets there.

The rest are on fresh pucks prepared the day before inoculation 6 days ago.

Pan Bisporus

I am really happy to see these doing well. They are bits of myceliated hpoo/coir substrate from a tub that wouldn't fruit. I am hoping that clean myc from these pucks will eventually lead to a fruiting culture. The ones with the light blue cap are from substrate dipped in h2o2. The last one is substrate that was not exposed to h2o2 (hydrogen peroxide). They seem to be doing equally well or maybe a slight lead to the dipped ones.

Subtropicalis Bell Cap

Dipped stipe pieces dropped on one side of the pucks doing well. One burrowing into the puck and the other crawling across the top.

Cube Golden Teacher

These and the rest are a myceliated millet grain transfer from a jar about to be spawned. I thought the jar looked a bit bacterial so I wanted to try cleaning a bit on a puck while spawning the rest.

Mexicana Chicon Nindo

These guys are beginning to take to the brf, but not nearly as fast as the cubes and pans. I don't know if it is because they are a bigger clump of grain and have a nutrition source that they are content to stick with, or a slower or less healthy culture, or they don't like brf pucks.

Pan Cyan Brooks

This has been the least aggressive pan that I have worked with but it is beginning to reach into the brf.

Pan Cyan BVI

I failed to take a clone from my first run of BVI so hoping to succeed with this culture to get it into long term storage this time.

For 4-5 year old spore syringes I'm surprised I have growth on 80% of my BRF jars and it's only been a week.
The jars I jabbed swabs into 5 days ago show no growth good or bad so at least those swabs weren't too dirty.

Nice success rate for old spore syringes.

I realize that it is a little late to ask now, but did you dip the swabs in sterile water before embedding them in the pucks? I think that helps, though it can work without it. Just more slowly.

That should help.

I haven't done this yet, so take it for what it is. The inside of the puck jar will frequently have condensed water on the sides or lid. Especially with fresh pucks. That water should be sterile. So in theory you could just soak up some of that condensation with the swab before burying it in the puck. I can't vouch for this because I just thought of it and haven't tried it. But could be a simple cheat to try next time I do up a batch. I believe I have some 4 year old PE swabs around here somewhere I can try this with.

Good idea, just one more way to save a step.

Also need to share a pic of my Malabar Coast jar, I've grown Malabar a few times
and it has always been aggressive.
A few years back I put a contaminated tub on my back porch and got 2 more flushes out of it.

I still haven't tried pf tek. Now that I've found vermiculite, I suppose I should give it a try to see how it works. Looks like I might need it in a rescue operation some day.
