Sparked my curiosity

The more grown out plate was the first dish I dropped spores to. The other two are transfers I've taken off the first dish.

I'm just curious as to why mycelium refuses to grow in that 1/4th of a spot? No biggie.. just trying to learn what may cause this odd occurence. Any opinions?

Contamination would be my bet.  That plate doesn’t look healthy.

Can I? I have a few plates I'd love to look at under a microscope. I also have all these spores that are listed "for microscopy only" that I need to do something with :wink:

God, I get frustrated with these invisible contaminations.  I don't completely doubt they exist I guess, but it's like a game of emperor's no clothes.

Is there really something there?

Perhaps it got that way due to sectoring?  It's just a theory.  Keep an eye on it and see if it fills in.  Have you been watching it and it has been that way for awhile without movement?

There is a bacteria blob at 3 o'clock on the plate nearest the camera.  That can be plainly seen.

Dude, I've grown a lot of plates and never ever seen that happen due to "sectoring". It has always ended up being a contam. Also, that fact that there is visible bacteria elsewhere on the plate kinda gives it a way. Even if the plate appeared completely clean I'd say it was something nasty occupying the space. BUT.... I've been wrong before.

I think I see some bacteria too in that plate on the left where the colony isn't growing out. You can see a sort of film on the surface. I've had plates like that where the main thing you notice is the sudden stalling of the mycelium and a reduced transparency of the agar. It's possible there was bacteria mixed up in the agar itself when you poured it.

Well I just took transfers of both of these plates as well. Slowly trying to grow out a fully clean culture.

Hopefully it works on my benefit. The agar was PC'd accordingly. So I hope I didn't waste all these plates/time on that.

I guess time will tell.
