Sometimes i do sometimes i Don't. If im making three transfers from one plate to three new ones i might not.
If im taking three transfers from the spore plate to three new plates i will flame so I do not cross contaminate genetics or possible contaminants

well said. It becomes common sense after some time and practice. If i know that my culture is clean and know that my scalpel is still clean, i don't take the time to re flame it. I took five transfers to new plates last night without flaming between.

It is true that a scalpel can have invisibly tiny myc fragments on it that can come off in the agar and grow out. For this reason, if you are taking from multiple different genetics and don't want to mix them up, it is important to flame. As a rule of thumb though, a noob should just flame between every transfer until they've gotten really good at sterile transfers.

By flaming every time when something does go wrong It'll be easier to trace back to where the problem originated..

Man, I thought I was going to get lynched for saying I didn’t flame every time.  really consider myself a noob, just want to be efficient not lazy.  I don’t think I would suggest noobs sterilizing every time cause in my mind flaming causes more hand movement which causes more air movement, and for me a warm blade melts the agar and you lose more ability to hold your wedge, and maybe it cools on the blade causing it to stick when you want to drop it.  And maybe I am over thinking it.
Edit, I wrote in the context of agar to grain or good isolates a2a.  Doing 1st or even second transfer I would suggest flaming.

I've been wondering about this, I also imagine that youd prolly have a much smaller available genetic material by using almost microscopic fragments to transfer.

Take the extra time, flame your tool, and let it cool.

Once you have confidence in all aspects of it, Tool sterility, deftness of movements, eliminating any unnecessary contact with your tool to anything other than your sample.

I still haven't tried the "poke". Maybe I'll give er a try but haven't found the need yet. I know it'll work though. I once did a series of transfer tests in which i just dragged the blade through the edge of a culture and then dragged it through a new plate. Those tests almost always grew out. I also did tests where i jest poked the tip of the scalpel into a culture and into a new plate. About 40 percent of the poke marks grew out.

You only need a few cells of mycelium  to colonize most things. depending.

I did the poke directly to grain took.around 28 days for 100% including shakes.

But i mean shit if you can knock a grain jar you can knock an lc. :shrug:

It's truly amazing. I'm starting to think that it's actually limitless. The other day I made SOOO much LC out of a nickle sized growth on agar and started thinking about what could really be achieved with one clean plate. Rough math...
1 poke per 1000 cc LC....
pokes available in plate.... ? let's say 100 even though it's way more.
100 pokes into 100 1000 cc LCs and your at 100,000 CCs of LC.
10 CCs per 4 quart bag of spawn and your at 10,000 bags
1/2 dry per bag over two flushes and you've got 5,000 lbs of magic.
5000 lbs from one plate. WHAT THE FUCK.

Yeah ive been making my a2g smaller and smaller.
I do 8 jars from one 60mm now.



It could be even more than that as well.
And faster. 1 lc.to 1000 lcs.

overexpansion  could become an issue maybe.
