Agaricus Bisporus Senescence and Sterility (Infertility?)
    #7971166 - 02/03/08 01:05 AM (12 years, 11 months ago)

I've been reading over some posts where someone will ask "can I clone a portobello from the grocery store?" and the general answer is something like "yes, but most likely it will suffer from senescence".
Why is this? Are the mushrooms we get at the store clones of clones?
Also, I took one out of the fridge that hadn't opened up yet and left it sitting out for a bit, until the cap began to open. I put it on tinfoil and left it overnight, but no spores have dropped onto the tin foil. Is it sterile? Whats up with it, I really would like to have some portobellos I can sell at the farmers market this summer...

they have to be mature for them to drop spores. More open than what you think.
best bet is to go to the grocery store and find a mature one and get a print from that.
Germinating the prints can be tricky. it helps to put a chunk of live mycelium on the plate also to help the spores germinate.
I am tired and I cant think of what its called and why.

Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

Commercial farms often buy spawn from spawn suppliers who do a long series of grain to grain transfers before finally shipping the product. The cell lines are so old by then they rarely make productive cultures. You'd be better off getting spore. Agaricus spores are white, so don't print on paper or you won't see them.

By white do you mean dark brown?

More of a light brown I guess.  Not white though, you're right.  I was brain farting on oyster spores, because I had just finished cleaning up another horrible mess from them. :fishsmack:

No members of Agaricus have light brown prints, they are all chocolate brown.

Yes, chocolate to very dark brown (sometimes appearing nearly black).

Looks kinda like light spore deposits, and very illuminated image.
I've never found any Agaricus to have tan-light brown spores, but maybe that's just my perception.

Whatever!  They're brown spores,  we can all sleep soundly now!

...Cool. Thanks guys, I guess my best bet would be to try to find a spore syringe online then. I'll get to searching...
Thanks again everyone!

Tahoe, I'm assuming this is something along the lines of what you were referring to?

"The stimulation of the germination of Agaricus bisporus spores by mycelium of the same species has been shown to be due to a volatile metabolite, diffusing into the culture medium and into the atmosphere. A wide range of other fungi has been found to affect A. bisporus spores in a similar manner. There was no evidence . that the stimulant was carbon dioxide. A seven-carbon olefin, isolated from air, which had been passed through cultures of A. bisporus mycelium and dried with phosphorus pentoxide, showed germination-stimulating activity but may have been produced by the action of this drying agent on some other metabolite from the mycelium. Volatile materials reported by other workers from A. bisporus mycelium and from Saccharomyces cereviseae were tested and, of these, iso-valeric acid and iso-amyl alcohol were found to stimulate spore germination of A. bisporus"

RR, I was reading a tek lately for woodlovers. In the tek, a bunch of woodchips are bleach water sterilized, and then a woodlover is liquified in a blender, and then sprayed onto the chips and left to colonize. I was thinking, taking a bunch of portobellos, blending them, and spraying them onto bleach or heat pasteurized straw. Do you think this would help, since then spores would alse be introduced to the straw instead of just using a tissue sample added to a LC, innoculating grains and then going to straw? I hope you peek at this thread again to see this.

I wouldn't try that with straw.

Bummer. Thanks anyway, I guess I'll be looking for that spore syringe then.
Cheers!
