Do we always Flame between transfer?

Is it standard to take a torch to your scalpel after every single transfer? For example if there’s 1 culture in a single dish and I want to take 3 transfers to 3 separate dishes flaming every time is not overkilll but is standard practice correct? All my work is performed in a flow hood

I often wipe down with alcohol soaked paper towel too, dipping might be more efficient I’ll have to try that

I spray everything with iso and flame like it's the only thing keeping me alive. Better safe than sorry and whatnot.

The dip gets any sticky agar off the scalpol and just adds that bit extra. I've been having a problem with my alcohol lamp not getting my scalpol red hot, I haven't had many issues but I think a torch is better.

I never make a transfer without the flame first and I never have broken my rules..

Wipe it to get crud off if needed because it's not helping sterility. If you can see any glowing of the tool in regular ambient light while flaming it then the metal temperature is already over 1000°F.

And yes, flame after each transfer.

Yes, flame it red hot between each transfer.  You will have a notable increase in contams otherwise.

I flame once and if I touch anything with the blade, and I don’t have problems.  And I’m sure that offends some people, but it works for me.

This post is strictly if you have a clean culture and good technique.

I use a fh and will do 5-7 transfers between flames.

If i switch cultures i reflame.

Whoa ok so here’s Bod saying no Flame until you switch cultures! Obviously having the clean culture and good technique to work with is crucial. I probably won’t attempt it when noccing up grain jars for awhile, but I’m going to start trying that with regular transfers. If I start getting random contams I’ll have to reconsider

It's about common sense.

If your flamed blade touches a sterile culture, and you take a cut and put to another sterile dish your blade is still sterile because everything it touched is sterile. Like van said, if you switch cultures you flame so you dont cross contaminate cultures.

However, from an educational and instructional standpoint you ALWAYS flame.

I had one transfer last week where I lifted my dish over the jar and knocked the wedge in after it fell apart while trying to get it on there. Maybe it was too hot, but I said if one is contamed I'll know it's that one. One did contam, also I think the next one.

Deftness in movements is key.

Sometimes i do sometimes i Don't. If im making three transfers from one plate to three new ones i might not.
If im taking three transfers from the spore plate to three new plates i will flame so I do not cross contaminate genetics or possible contaminants

Watch my sab videos on agar work. I cut corners with flaming. Some people noticed. I did it so some people would catch on to the common sense part of it not the robotic following advice because I can't make my own decisions stuff.

A good method is also to mark any jars where you notice a flaw in your technique, that way if you end up with problems you can easily reference back to the transfer and identify your vectors
