Strain Isolation

Looking for further information on strain isolation.
I'm grappling with the basics, I've watched RR's video and am looking for more sources of reading.
If you've got anything in mind that will help give me more direction and instruction let me know.
Flipping through the searches in the meantime!

Sectors seem very difficult to actually identify.. but after seeing this second example its solidified a bit.

Sector my pecktor lieutenant protector. Thanks for moving this over though really.
I'm going to turn this thread into me taking peoples plate pictures and trying to identify sectors labeling them out in MS paint when i've got the free time to do so and I'm not playing moron somewhere.
If anybody else feels like it go ahead and sector those pickles and share them here and we can figure out if what you drew was a proper sector selector or some bullshit wutermelon juice.
It'll be like a game of darts at the bar but we can learn stuff.

More ppl will reply here. Some people have documented their isolation work. Its ridiculous work. Better to clone or streak MS extremely dilute and test those cultures

You won't see much sectoring until you've made a few transfers. The reason is that from multispore inoculation you have thousands of sectors piled up on top of each other so it just looks fuzzy.

Take a few tiny pieces of healthy mycelium from the very leading edge of the growth and transfer to new plates.  Repeat when the growth is less than halfway filling the plate.  After the second transfer, you should start to see sectoring.  Take tiny bits when transferring; around the size of a grain of rice.

If you're still around, I've yet to manage to get my refrigerated cultures to remain clear with no condensation forming after storage.
My temperatures are set to 36 degrees with a 2 degree fluctuation. Its a dedicated refrigerator in my lab space so it doesn't get opened often.
Awhile back I had asked around here on the forum and it was summed up to being just how it is but I'm not too sure that is accurate.

From one of the previous posts I seen you leave around here you said storing it in an insulated box can help protect the cultures from the fluctuations in temperature and if I remember right, protect it from forming that condensation.
Should I just go ahead and start sticking my new cultures in an insulated lunch box before putting them in storage? I haven't gotten a chance to test this out just yet being I've not made up anything new since touching on that in the past.

Yeh, improved consistency. Given identical conditions, a monoculture will perform identically between different grows. This is great for knowing when crops will be harvestable, so you can schedule deliveries to your customers.

But, getting a good monoculture is a laborious process. You have to do many transfers, and test all possibilities. Very few commercial growers develop their own monocultures, but rather buy them from specialists, who have teams of people whose only job it is to isolate and test.

Isolating from MS is dumb. Isolating from a clone is better. At least with a clone you know you have something that produces fruit, and can verify its fruiting characteristics before putting much isolation effort into it.

I put a clone to one plate, made one transfer and it's looking really good off of that one transfer. Anyway, what benefits do you get from isolating a clone? I mean, for the most part aren't you going to get the genes from what you've cloned regardless?

Transfers just for fun to "isolate" more?

Clones are a few sets of genetics working together. So u'll still get some variance within the tub. A true isolate will produce exactly the same every time because it's only 1 set of genes. But there's a thread on here where dude tried for a long time to make an isolate and never really got there. Seems like wasted effort unless it's just a side project and u have the patience. U can store clones on a slant if u have a good clone.

But yea, I guess it's just for fun. Science!

Ah, makes sense. Yeah I'm good with just taking a clone and making sure its clean before putting it to grain. Maybe more down the road I'll play with things like that. But for now, I'm in it for the yields!

2. Reduced variability: In your MS tub you may have millions of individuals all growing in,  amongst and around each other.  MS fruit are multiculture, but have far fewer than the agar you transferred from.

Isolating from a clone is much quicker than isolating from a print.

You could still end up with a dud, though. It may happen that a non fruiter, or a mutant ran along with a fantastic fruiter, and you isolated that. Either way, isolating is a great way to practice and improve your transfer technique.

In anycase, most monocultures aren't that great.

This year I'm exploring a Thai clone that I did end of last year. I took 8 clones from a single cluster, and cleaned them up through December and January. I didn't isolate, beyond what isolation happened during clean up. There was some sectoring at the final plate, but not as clearly delineated as I've seen from other's pictures. Nothing fantastic yet.

Alright. Well regardless, I'm pretty happy to put this clone to some grain. This other shoebox I have has a cluster of like 5 with a juicy one right in the middle. I've already found my next baby I'm going to put that work on hehe.
