Hello,

I started with agar, got PDA mix nearby so went with it. I used 150 ml water with 1 Tbs PDA. Can someone here confirm this whether it is right, it solidify alright after pouring , although i faced condensation.

I inoculated 15 minutes after pouring, was  not sure how long to wait and whther it was too hot to inoculate.

Another issue was i used a little bit thicker wire to make my loop , so still doubting wthether it was too hot and whther i killed spores during the process.

Today it is 3 days after inoculating 3 plates , 1 shows clear contamination ( will later post pic to confirm this too) , 2nd plate shows nothing , 3 rd one which shows a small dot which looks (hopefully) like myc .

I have 3 uninoculated plates in fridge which i plan to trf from these plates.

Another prob i faced was after sterilizing loop when i dip in agar for cooling , the area became black . I used denatured alchohol lamp, still dont know why it happened.

SAB work was a bit of mess , as it was my first time.

Also i want to know should i keep inoculated plates in a air tight box or ziplock bags?

Clarifications will be helpful.

Yes the agar is suppose to stiffen after you pour it. I stack my dishes 20 high and let them cool like that. It helps with condensation

15 mins is probably enough time, if when you poured you could handle the agar.

I use a paper clip bent into a loop, not sure what a standard size is. But as long as you cooled it on a dish first and it stopped hissing, you should be fine

Germination can take 3 days to 3 weeks.

Its probably turned black from the smoke. As long as nothing is growing don't worry.

You should practice the movements in your SAB, to get a better feel. I like to record myself from above, so I can look and see what I did wrong

I wrap my dishes with plastic wrap and store them in my SAB in my closet. Mostly so they are out of the way. I'm sure you could just leave them on a shelf or whatever. But you should wrap them either with parafilm or plastic wrap

Hi,

I had inoculated with 3 plates with Agar. I need advice whether these are contaminants or myc . I used micropore tape, so the dishes look slopy.

Plate 1:



This looks like clear contamination on Top and myc on bottom.

Plate 2:  I have taken pic from 2 diff angles.





I think myc is growing on left and right side circular, what is it? contamination?

Plate 3: Again from 2 diff shots.





Here too there is a circular dark growth on top and rest myc ?

Its my first time , so doing only guess work , members with experience pls comment.

Also when should i trf to new plates , its 4 days since inoculation from spore print.

yes plate 3 looks like some myc growing , but i want to know what is on plate 2 left side, they are growing vigorously.

I think you have what you are looking for in those plates but unfortunately all are contaminated with mold and bacteria, reassess your technique in the sab, make sure all surfaces inside the box are wetted down with soapy water or even just water then leave everything set up for 20min or so to let contaminant spores settle before starting work, open dishes for the least amount of time possible and don't let anything pass over the open dish apart from the scalpel or loop, make only methodical movements in the box with your hands, not jerky or too quick.

Plate 1 or 3, looks like it has some healthy mycelium. You need to transfer away from contaminants. I can see bacteria and probably mold. Dish 2 I would just toss.

I would take a rice sized, or smaller peice from the leading edge of healthy growth for the transfers. I would do it soon. Then do at least one more transfer to ensure you have a clean culture.

Also, if you can't find parafilm to wrap your dishes. You can just use food grade plastic wrap.

Have you smelled the mushrooms were u got the spores from before? You could clean a plate a couple times on the limited choices an smell it but would just restart an norrow your vectors.



Move you hands really slow like a chameleon, the videos on here of people doing it are to fast.
Barely open it no more than u need very slow so it doesnt suck air in an flame everything that goes over your media.
Also let or make condensation go away before using them, thats were alot of infections around the edge come from an the excess water spreads it when its moved around. You wanna leave plates be untill your sure nothing else is in there(spore plates are always dirty.)

Nice tips , i inoculated immediately, there was condensation, my movements in sab was far from methodical and am not too sure about my PC tek. So my first attempt at best was  a learning exp only.

And i got my spores from a vendor, and i didnt understand the smell part.

Each mushrooms mycilium usually has a unique smell, like reishi myc smells like reishi fruit in the same way cube myc smell like fresh shrooms.


If your unsure if it white mold or myc grow a plate of it out an smell it. But you would need to know what your smelling for...

Never smell a plate u know has nasty shit in it cause you can make yourself sick only for plates were your pretty sure but just need to know.

hey all,

just an update, me still struggling with white mold contamination in agar plates, even after 4 trf i am left with only 1 agar plate which is clean.
Out of around 25 plates i managed only 1 which was clean. This even after taking utmost care what i learned here.

Also i did trial inoculation of wheat grain . Its 15 days now . But i am not 100 % sure its myc or white mold . Initially i confused mold for myc in agar plates, and it took me some time to know the diff.

Now i need opinion on my grain jar.Pls look at the pic and pls comment.

Also pls direct me to some good threads on contamination in jars, specially contamination which looks similar to myc.

Its hard to tell from that pic looks like you used an Instagram filter. Id say when its 100% crack her open and sniff it out. If it smells like mushrooms spawn it.

It is most likely myc. Looks fine other than a bit fuzzy for not yet fully colonized spawn. Like bph said above; once it's finished open it and smell. I'm not sure if you know what fresh mushrooms smell like. It's a musty, earthy smell; that as you continue in the hobby you begin to associate with success and hence begin to love the smell. I'm not sure off hand what white mold would smell like, but I doubt it would smell good.

Was this inoculated from spores or from that agar?

the above jar had turned out to be contaminated, i disposed it, another jar which colonised slowely (took 1 month) is now spawned to coco coir in a small plastic container (since i had only 1 jar of spawn) and kept inside a plastic bag.

I didnt find any tek here for this, but i saw a video in youtube and went ahead with it.

At this stage i need a few clarifications:

1- I kept the box in a dark place, opening only for spraying , will the light cause any prob?And after what stage should i introduce lighing?

2- What about CO2 , whether it should be high or should i allow for more fresh air exchange?

3- What humidity should i maintain?

Also pls refer to any tek where colonising and fruiting is done in a plastic bag.
