1st generation - confused

I am confused about maintaining spawn and cultures. As I understand, once transferred to a new medium, it is considered in the next generation. How does one perpetually maintain 1st generation cultures if one keeps cutting out pieces from the first generation culture, soon, there will no longer be a first generation. Where do they/does one obtain from to perpetually have a first generation?
Initially I thought one could do a tissue culture of their own mushroom growth, but then I read that would just make it the next successive generation from whatever spawn generation it was grown from. Is this correct?

Cold storage of cultures.
If you make 10 masters on dishes or slants. You can pull from each one dozens of times before its "gone"
Each pull can get you going for months to years before you need to go back to master stock.

I’ve been reading up on differences between the Petrie dish and slants and sounds like slants help to maintain the master culture best/longevity? Dip into that to make a few 1st/2nd gen Petrie dishes and I guess use that to multiply from?


What is the best way to go from Petrie dish to inoculating 6 lb bags of grain spawn? Should I make a few smaller 1 lb jars, then transfer that to the 6 pounders(because there is such a large volume to inoculate)? Or should I go directly from Petrie dishes to the large 6 lb bags?

Think of it as a growing/expanding organism.

you can take one tissue sample from your slant, grow it on agar, then make ten dishes from that one. When all ten are ready you can then half or quarter them and make twenty jars of grain, which can then make twenty bulk bags.

When the myc gets "tired" you can get more tissue from the slant and start over.
