when you are making slants with a small stick in them, do you cook the agar before you pour it into the test tubes and then sterilize the tubes with the stick in them or do you just mix up the agar and pour it straight into the test tubes and then let the agar cook in the test tube?

you cook it, to jell it up. then pour.

The way I've seen it done, you sterilize while agar is in the tubes, liquefied.

No, it won't burn, 15 to 20 minutes at 15 psi is all it needs to cook in the tubes.

I soak the wood tongue depressors for an hour in water. I then mix up the dry ingredients for the agar first, and then add water, stirring until all the powder disolves. Pour into test tubes, insert wood, screw the lid on tight, and sterilize 20 minutes at 15psi. After removing from the PC, lay the tubes to solidify at an angle, using whatever is at your disposal to obtain a 'slant'.

If you used food grade agar it is usualy not milled as fine as laboratory grade. The food grad agar, if not cooked first tends to drop out of suspension and you get agar that is heavier, thicker at the base of the tube, with laboratory grade agar this is not a problem.

RogerRabbit good point about soaking the wood first.

you screw the caps on tight before you cook? you dont have a problem with the tubes breaking?

Correct. Screw the lids on tight. Nothing breaks because there's no pressure differential between inside the test tube and outside. As the PC heats, the pressure inside the tubes follows that of the steam surrounding them. I've never broken a test tube this way.

Putting sticks in the slant greatly extends the life of the slant. The mycelium will grow into the stick provides a better environment than just a plain agar slant.

I'll have to try that, thanks.
I'm gonna PC some tooth picks and stick them in some PF jars or throw them into some agar plates.
I hate transferring little bits of agar, they never behave...tooth picks would be easier.

Tooth picks might work but I recommend you get something a bit bigger like the stir sticks you get from starbucks.

  • Break down your sticks to fit your slant tubes.

  • Soak the sticks first in water for ½ hour to keep them from floating up in the slant when the agar is added.

  • Mix up some agar and fill the tubes about ½ to 2/3 full

  • Put the caps on and wipe clean.

  • Sterilize for 20 min at 15psi and then let cool

  • Remove from cooker and place at an angle and let finish cooling

  • The mycelium will move into the wood and colonize the sticks.

This is a good way when you are going to store a slant for an extended period of time.

I wonder if they'd go for bamboo, chopsticks are pretty meaty.
I got a bag of those craft sticks..like Popsicle sticks, I'll try those too.
