Amazon or Ebay, I have a couple 250's coming in. A buddy got a 1000ml due to the shipper messing up. He was refunded and told he could keep it, he gave it to me, its huge...lol I still need to get a 500, I couldn't beat the price on the 250's so I scooped em up. Just search, 500ml pyrex media bottle. About the cheapest I've found em. Hope that helps.

Duran is the original and best. You can play basketball with them - but don't. They aren't too expensive, unless you get ripped off. FYI: If you order the bottles with caps, they usually will include the pouring rings. There are an assortment of caps, some which have a semi-permeable membrane which will allow for GE, but polyfill stuffed in a drilled hole does the job.

Don't fill them more than half with agar, and loosen the cap slightly when heating or sterilizing. But, yeh, 500 ml is great for agar prep! I use that size just for agar, and prepped agar will store for weeks in the fridge. I use 250ml for LC.

Best place I've found them is a local distributor, requiring a Google search. You should have one, and the guys should only happy to help get you what you need.

Yeah don’t even worry about drilling any holes for the cap for GE. Not necessary whatsoever.

I would still put a filter in it. As things cool in a jar they draw in air. You want that air filtered.

Is this commonplace? I have been using Bods Comprehensive Agar tek with an unmodified lid for quite some time now without fail.

Well I’ve always done it that way, which is why I am trying to fathom the need for a filter since I was just given conflictive information from a TC. I wanted to be more informed. I mean I get it, seems more like an extra measure to ensure a sterile medium, but IME, haven’t had any issues just loosening the cap ever so slightly.

Just for extra safety maybe :shrug:

I'll admit I've reached in and grabbed agar bottles out of my pc and forgot to tighten the lid back down before I took the foil off...It hasn't fucked me yet, but I've definitely cussed myself for it :facepalm:

Haha, that’s always the first thing I make sure to do! I could see where that could be an issue though trying to grab it by a loose cap.

I think you hit the nail on the head. Having the filter would ensure that the cool air getting withdrawn into the medium would be less contam prone than just leaving it open. I just didn’t know if this was something a lot of people did or just a few. It makes sense, I just know how much I love to swirl my agar around in the bottle and the thought of it touching any kind of filter makes me cringe.

IDK how many other people use filters in that type of bottle but I will always use a filter, no matter what.

I’ll have to get another cap and try it out for myself rather than just knocking it, because I’m sure it’s more ideal than the unmodified lid knowing that the cool air is going to inevitably be sucked in. Wouldn’t hurt to say the very least. Thanks for the banter, needed this!

Ive poured over 2,000 plates with the bottle as is no filter in my home. And tens of thousands of them with the same bottle for work.

I believe you but it still scares me. I wouldn't try it. Drilling a hole and slapping on a filter are easy and cheap enough that I'll still do it.

Since I purchased new 500ml Media bottles I no longer use a filter....my old setup I drilled an 1/8 hole used polyfil, but now I just do the loosen cap method...Works even when I forget and take the foil off prematurely :lol:

I don't like a filter on agar prep jars simply because you lose moisture when storing, and you can't shake it up when heating for a pour. Yes, there is a risk, but honestly it's minimal. 18 ml of air will be sucked in as a half full 500 ml jar cools from 100C to 75C.

I'll just trust that since these bottles are literally made for pouring agar. Yea..

As soon as your PC hits 0 open it and tighten the cap. Loosen it in your SAB or in front of a hood.

I ordered a couple of these bottles today. The 500 ml media bottles. I’ve always heard don’t fill them up to the 500 ml capacity, and to only make about 250 ml of agar per 500 ml bottle. I guess people are having boil over problems?

I was planning on making about 40 plates so I probably need about 1000ml agar. It’s why I bought two bottles. I was just going to fill those bitches up.
