agar questions

Hi I have a few (SDA)agar plates inoculated from  prints 1 week ago and there are spots on the plates now.The strains are Azur,Ps.cyanscens and Tampa .By Tampa it's very obvious which spot to select for tranfer but the woodlovers have got a lot of yellow spots and a few white spots and they aren't cottony and aggressive,anyway.And by Ps cyanscens there is a late myce. appearing on the plate which is cottony but fluffy.There are also yellow spots with green colours .So I thought I tranfer the white spots thinkin they're more healthy than yellow ones and yellow green ones :wink: and the fluffy white myce right?I'm not sure so please help me guys what to do?

yep, do an agar transfer with the healthy looking stuff. since theres now myc. you can use an antibacterial/H202 in the agar and that should help with contams.

also, one of the biggest agar contams i was having was the lower few that were sitting directly on the floor of the incubator had tons of bacteria. i propped them up on some styrafoam and havent had any problems since, seems its too warm if they're touching the floor.

I use a very dilute amount 1cc of peroxide into the 500 ml of agar and it seems to keep out the baddies.
You wouldn't want to use too much peroxide since it will stall and possibly kill a very young mycelium network.

The only thing I'm not sure about is:
I took a scalpel and cut the part of mcye I want with the agar under it.(I cut the nutrient(with mcye) and not the myce itself right?)and
Should I use the H2O2 in the new plate that I'm gonna transfer the myce or should I spray the old dish first to clean the myce part I want and then transfer it to a new dish or both.Sorry about my english.

You will want to use peroxidated agar for your new plates, the plates you are transferring to. It's fine to transfer the agar wedge (mycelium and agar), it's hard not to.
It sounds like bacterial contam so you should be fine doing this but if there is a fungal contam I usually try to only grab mycelium (no agar) from the original plate. The reason being, if the original agar is brought over into the new plate and there are other fungi spores around the original agar acts as a medium in which the contaminate spores can germinate. It is then able to spread into the peroxidated agar as this fungi most likely has peroxidases, same reason the wanted mycelium can grow in it. If you only transfer mycelium only the mycelium transfered will grow. spores should not germinate.
But it sounds like bacteria so you should be fine with what you said.
