mycelium died after shaking ?

Hello and thank you for all the answers to my question about the explosions in my PC, I manage a little better now. now I have another problem : I inoculated 5 jars of rye with mycelium grown on PDA substrate, and after 5 days they looked really good and colonized quickly (around 29 celsius)so I shook them as recommended in The Mushroom Cultivator, but now they seem dead and the mycelium doesn't grow anymore. these jars were sealed (I live in a small island and I don't have access to such thing as synthetic filter discs), is it the problem ? I don't think it's contamination from outside because I work in a glove box with HEPA filter and follow sterile procedures. Endospores maybe ? if it's the case will "tyndallisation" help ?

if you are using grains thee is a greater risk of contaminatio with out autoclaving/pressure cooking... do the kernels have a greasy apearance or is there water that had collected in the bottom of the jars... these are signs of bacterial contaminants... it sometimes sakes a few days for mycelium to recover from shaking and also if the baccilus is present then they may not recolonize at all... best to wait a week or two and see how things progress

Can?t you get some polyfill in the island you live ? Polyfill is sold as pillows filling.

Use polyfill.  That is what I used even though I probably could have gotten some Tyvek.  My quarts of WBS are moving along nicely.  :smile:  Good luck.

thank you all I will try to find polyfill. The thing is with my jars, they don't show any trace of contamination at all, they just don't grow anymore after shaking. I think now it is the total lack of air exchange. Thank you for helping ! Pedro

If you find polyfill hard to source try micropore tape, works great and they sell it at most pharmacies.

if you live in the united states, or a location the USPS services you can have them send you free tyvek envelopes, which work as great and free filter disks! i ordered enough for the rest of my life(20 packs of 10), they arrived 1 week later.
