Liquid Innoc. (again)

Well, it's been talked about at least 3 times here this week, and I did a comprehensive search, yet I am still not comfortable going forth with my first liquid Innoc. tonight. Primarily, what is the best liquid innoculate for: 1) potency 2) Ease of use What is the best Tek, and what are the best products (brand) to use? I intend to make enough liquid innoculate to do 6 QT WBS jars, and 25 1/2 pint, MMGG jars, I always use ground Millet instead of BRF in these. I really welcome any and all input on this, cause I want to start doing this process correctly, right from the outset. My thanks to the many Shroomerites who have answered these questions for me in the past, and continue to do so to this day.

PDY is the best, IMO, but karo will work fine and it's easier to use. Get some septum plugs to put in the lids of jars or something unless you're going to be working in a glovebox. Cracking open the lid will probably result in contamination -- if you absolutely must just crack it a bit in back of a flame.

Coolness, just straight 100% Karo syrup right out of the bottle?
And I was going to use a 1/2 pint jar with a small filter, and a self-healing silicone spot for the injection point. This should keep me from having to expose the contents to contams.
Would anyone care to link me to their favorite Teks, also?

Hi JetBlack, No not straight 100% karo... Here is the formula. 75-100 cc's of sterile water. (I use 75) 1 tsp of karo or honey. 1/2 pint jar is fine. PC for 20 minutes, no more than that! And thats it..I close it up tight rubber seal down. let cool then take to clovebox for inoculating. Ive never had contams.
Karo is crystal clear and easy to see into. (checking growth and contams) Honey is cloudy and not easy to see into but twice as fast as karo imho. Do not let it grow out very far, You will be tempted to let it get thick as you can but it will be a big mistake when you try to suck it back up into the syringe. a couple pieces of broken glass in the jar before you start will help agitate it and break it up in pieces.

Right! Glass works wonders, especially if the mycelium is really thick. Only....

Watch out -- you want to shake the jars like once or twice a day for airation, so a filter which is porous will leak the liquid. If you only fill the jar like 25% of the way, though, it should have all the air it needs -- just seal and shake every day! Airtight is the best to prevent contamination, anyway -- like I said, I work without a glovebox, so it could be a problem.

okay, what about a larger container?
can I keep it in my incubator at 82-86F?
I need to shake it twice a day? See the funny thing is, afof is really doing these, and he can't be trusted to remember anything.

Size doesn't matter
No, really -- any size container will work, but only fill it max 25%. I use 500 mL bottles and run like 200 mL in each (I guess this is a bit over 25% but whatever.) 200mL = way too much for me, anyway -- that's 20 10cc syringes. x 5 bottles = 100 10cc syringes....
I would definately shake them every day, though -- you want O2 to dissolve into the liquid.
Also -- if you're using air-tight containers you might want to let them cool inside the P.C. before opening and sealing them -- I didn't let them cool and it formed a vacuum inside the jars.
Temp isn't critical, but 80-85 is the best -- I'm sure 70 would work too but slower.

Nice thread, good tips.
I've tried liquid innoc with malt extract, the kind used for beer brewing. However, this tek is, IMHO, very shitty. I've only had one jar out of 4 come out with growth, and it was hit with contams right after I drew the first few syringes. And it was really cloudy so hard to tell what was going on.
I'm going to try the karo tek tonite. Sounds a bit more promising.

Sweet ass. I will keep it in the incubator, that's what it's for anyway.
I will use a QT jar 1/3 full. It will have a silicone self-seal injection point, and a filter to allow for air-exchange.
Do I have to shake, shake? Or can I swirl it around real fast, which would keep the filter dry?
....and to get them started I just empty the contents of a retail spore syringe right into it?

What do you use for a filter? There seems to be a problem with my filters getting wet.

I use a material kinda like poly-fill, but it comes it roles, and is easier to cut into flat even squares.
That is why I would prefer to swirl than shake.

Without a filter, how would I get air exchange, and how would I be 100% sure to prevent the vacum effect?
and 2CC, are you sure that's enough for 1/3 a QT of solution?

There should be plenty of air in the jar, as long as you fill it 25% of the way, or less, and shake once or twice (better) every day. The air will dissolve into the media.
I don't think you have to worry too much about a vacuum seal -- I was using special narrow-neck jars, but to be sure just let them cool INSIDE THE PRESSURE COOKER and then open it and tighten the lids after everything has cooled. That should do it, I would think.
2cc's should be plenty.

Thank you kind sir. I will use, no more than 2CC's which is actually good news to these ears.
I still think I may put a small air filter on it, just because I have a knack for growing mycelium VERY slowly.

For liquid Innoc, I don't use a filter. Just 1 small hole in the lid that has silicone seal on it, for a self-healing injection point.

Your getting the idea jet! no filters at all. If you have gb or a flow hood, you can use a lid with no holes at all. just as long as you can crack it to shoot the spores in an aseptic enviroment it should be fine. Im a big fan of adding several cc's of peroxide to liquid jars after the spores have sprouted. It seems to speed up the growth and keep things nice and clean.
