Grain vs. WBS
    #2196182 - 12/22/03 07:05 PM (13 years, 11 months ago)

Would anyone happen to know if there is any difference between Rye grain and WBS as far as their susceptibility to contamination?

Does either take more or less sterilization to reach the same goal?

And finally could someone possibly provide a good water ratio for Fractional sterilization for different substrates? (specifically WBS and Rye)

Thanks everyone any help is appreciated
Trkgsy

I will never use rye grain again since using WBS.

Simmer for 1 hour, drain, and load jars, Pressure cook. Far more points of inoculation. the simmering really takes care of any lumping problems that occur without the simmering.

I am a convert!!!

How about fractional? (no cooker as of yet)  :frown:
If fractional does work with WBS how about that water ratio?

if you simmer. or presoak for 24 hours then drain completly you dont need to worry about ratios, the grains will have absorbed the correct amount of water.
takes out all the guess work.

When you say after soaking and draining that the grains will have already absorbed the proper amount of water, is that reffering to WBS or Rye or both?
I figure you meant just for WBS, since in my short Mycological expedition I have found Rye to be a tempermental substrate.

thanks

Yea buddy:)

Rye is ok for cubensis...and I guess that is what we are talking about.
Birdseed is better....especially for pans:)

Teonan said it....do it just like that.

A little head start would be to use an agar wedge.

Remember to use the best rhizo from that wedge.

I don't know how effective fractional sterilization would be for grains. Definetly give it a try. Simmer the grains first, both rye grain or WBS.

If you save money for anything in this hobby, a pressure cooker is it.

it works that way with all grains, some dont need the simmering but it will help with germination of some of the endospores that may reside under the hulls of most grains. popcorn is a good example, I simmer it until it splits and allow it to rest for a day, drain it for an hour and PC it for 45 minutes. not a problem yet.

Just a thought here, but does Rye have the same nutrient reserve that WBS has? Does this matter?

I bought a pressure cooker today but it's used and old, looks to be in good shape but no gasket. I posted about it today. I may still be forced to try Fractional Sterilization if this cooker is not safe, and until I can get a cooker.

I would like to do agar work ralphster, but it is a little above my head at the moment. I have read up on it somewhat and it is quite entriguing. I need to find a good source for agar, and spores too really.

Thanks alot for all the great help

Trkgsy

hey tricksy. don't worry about the nutrient content, as wbs is mostly millet and many here on the boards reckon millet is the dog's bollocks!

Teonan, i thought your 1 hour simmer was a little long, but i just tried it - obviously your wbs is different to mine, as mine was starting to burst after 40 mins. After pcing the grain was all stuck together like a pf cake!

good luck with the wbs - it does indeed rock  and merry crimbo
