making liquid inoculum

I was wondering if anyone has found a highly efficient method for making batches of liquid inoculum from agar or liquid cultures? I currently use an autoclavable blender, but get really tired of having to sterilize the blender top in between each use, when I am trying to make inoculum for a whole set of different species, in a short space of time.

Are there perhaps blenders available with disposable (or at least v. cheap and autoclavable) container tops, or maybe a hand-held mixer with disposable blade attachments?

I know there are very expensive mixer units that can blend multiple samples within sterile compartments, but I cannot find any cheaper options such as I've just described in any of my lab supply catalogues.

I can't just fragment liquid cultures by shaking or magnetic mixing using beads and broken glass, etc., because some of the cultures I'm working with are too tenacious/fibrous.

Any help'd be much appreciated.

I had a very good success using simple jars filled with water in combination with this



The blender rod is disinfected in alcohol, sterilized water and in 1% peroxide solution.

sometimes one of the simplest things works the best.i personally would never have thought of a mixer ! simple and easy, and easy to sterilize.

thanks very much Anno! I'll try one of these out. It had occurred to me to use one, but I hadn't thought about the possibilities of rapid in-between use sterilization. I'll try washing off debris, then soaking in ethanol, rinsing with sterile water, and then a quick blast of UV just to be sure. thanks again

Blender top? You *do* know that a regular mouth canning jar will screw onto the blender base, don't you? You could go to walmart, pick up several blender bases, and put each one on a regular mouth jar.

I *do* know that, in theory... but unfortunately i can't find an equivalent system here in the UK. My jars don't fit any base I can find. I think this easily cleaned hand-held mixer might be a better option anyway

Anything you throw in the jar can mix it up (glass beads, broken glass, etc.) and you can put a septum plug in the jar to make a self-healing injection site.

Someone just told me of a rubber seal that fits under the cover of a mason jar -- anyone heard of this?? Apparently it's available on the market....

so if tonight someone decided to Blend a portion if not the whole almost mature cubensis fruitbody into a mixer that had been sterilized along with a little water for transport assuming it sterile too, and was sucked into a sterile syringe underneathe the surface of the mix to avoid sucking air contaminants; That would work or doers it need modifications?

no u need a nutritional liquid medium(not just water) for a mushroom culture to grow on.. then u take the mycelium growth and suck it into a syringe..

You can mix any sterile, live tissue from any part of the fungus with water to make inoculum. With a nutritional broth it will actually grow, too.
