Help with growing from store-bought portobellos
    #21211601 -

Sup fellas, recently tried to make a spore print with baby portobellos, and do spore print to grain with what seems like no luck but no contams to be seen. I'm a dingus when it comes to collecting live culture or spores so I'm seeking advice from the elders

I have a few portobellos with mycelium growth at the bottom of the stipe in my refrigerator. How should go about getting some running mycelium or collecting spores the right way
Links are welcome!

Agar is the answer.  Check out the write-up in my sig.

I'll definitely give agar a try in the near future, but my question still stands. What procedures do you take to start growing from a store-bought portobello mushroom? I'll do more research on portobellos

You'd put either spores or tissue to agar, and then to grain, and then to a bulk substrate (preferably mushroom compost) and finish with a casing.

Depending on how much time I had, I'd make my own mushroom compost like Agar used to.

I'd just do a print to agar. just buy one break or cut the stem off and print onto foil. I just finished cutting some tissue from some crimini stems into wbs paste Won't know for a few days if they will take. But saved the caps to foil hoping to get a print or 2 the veils were broken so hopefully I'll have a few half assed prints in the A.M. I'd suggest looking into this. http://www.shroomery.org/forums/showflat.php/Number/19208976 Super simple and cheap too, just finnished my PCing the agar gonna get some Noced up tommorow.

You've all been very helpful, but I can't really afford to buy agar or petri dishes at the moment, could I just break off the stipe with mycelium growing on it and dump it in some sterilized WBS? I've tried to make a spore print and scrape into the WBS but I haven't see any growth and it's been at least 7-9 days since I knocked it up

Agar isn't so expensive. You can find at most asian grocery stores for 2 dollars for 25g.(I think this is enough for a liter or so which is a lot of agar plates) I use half or quarter pint jars as agar plates. they are less than $1 each generally.

I believe that Agaricus spores are quite difficult to germinate on agar. I would try to take a tissue sample on agar from a fresh mushroom. Preferably near the top/cap. To do so, clean the mushroom well, rip it into inside a still air box (you can build one, just cut arm holes in a clear plastic bin and flip it over) and then cut out a piece of the cap from the ripped section and place it onto agar. This way you wont drag bacteria from the surface inward as you would if you were to cut into it straightaway. I hope this helps.

You could certainly try... Let us know how it turns out if you do. Be aware it may likely contaminate if done like this though.

could try putting a slice of the stipe in a cardboard burrito. you soak cardboard in water until its saturated, then let it dry til its not dripping anymore. then you peel apart the corrugations and lay it flat on a surface. put the pieces of stipe inside and roll it up. then, put it in a plastic bag. it might work. it might not. it works for oysters but agaricus eats compost, so IDK. if you put the stipe in sterilized wbs, it will contaminate.

leschampignons has the right idea. you can use mason jars instead of petri dishes. or plastic tupperware containers called mini-rounds, which are autoclavable. you can mix your agar and sterilize it in the mini-round so you don't have to pour it. much easier for beginners. then, when its cool, put a piece of the stipe on it and it should grow out. cut the stipe in half in a sterile space and put some interior tissue on the agar. the outside has bacteria on it. i would do 5 or 6 and likely one or two will not contaminate. then you put the agar in wbs. its a little more work but not much. and it will safe frustration.

what you need: agar (asian food store), malt extract (brew store) or malt syrup (grocery), nutritional yeast (health food store), gram scale to weigh ingredients, mini-round tupperwares (walmart or grocery), pressure cooker (sounds like you have one if you sterilized grains), scalpel or exacto knife (regular knife will work if sharp) and tweezers. probably costs less than $12 assuming you have the pressure cooker
(edit: you can even boil potatoes and use the starchy water in place of the malt)

you might want to consider starting with oyster mushrooms, too. even i dont know how to grow agaricus yet...its a secondary decomposer, so its a little different to grow than most edibles

If you want to grow them, you need compost or manure. Even for agar, use manure tea in place of water. And you need agar for storing and inoculating substrates, without it there is no use of having a culture because you can't work with it. You would be better off then just blending few shrooms and mixing it in compost and hoping for the best. Good luck anyway

And for agar, no need for expensive extracts, just boil around 20-50 grams rice for every 100 ml. And use that for agar and don't forget to soak some manure, for agaricus yeast in the mix would be usefull too

Last edit: and for tools use disposable srynge needles as they come sterile and cut up agar quite nicely. A good start untill you decide to go serious.

One of the problems is that i have no idea how much skill or equipment you have, but...
Of course you can do this.
You would need to pressure cook or boil your grains 8 hours to sterilize them, you would need to work in sterile area when transferring sterile piece of tissue from the middle of the stem and you would have to know exactly what your doin.
On the other side, something similar can be done with oysters without pressure cooker or sterile area, because oysters are much more agressive. There is youtube video about oyster growing, just letme give you one last hint, dont belive everything guys in this videos say.

Agar is not needed tho it is essential media in mycology.

I work with a still air cardboard box, it's not air tight but I make sure to spray lysol when appropriate and gloves, a 16QPC and ofc jars but I'll try to pc some grains and try it out in the future also will post results might try the needle through the stem tek, seems plausible.
