cloning store bought mushrooms

Just got all the agar supplies. Brand new to agar and edibles, but looking foward to it. I think I have seen this posted before, but if I understand correctly, a fresh storebought mushroom could be sliced with a sterile scalpel to remove sterile tissue sample and innoc. the agar dish with that? Is this correct. I want to start with oyster since that sounds like the easiest, and straw is easily accesible. could a dry mushroom be re'hydrated and take a sample from the middle or would the water then carry all contaminants from the outside? If anyone has any extra culture's they would like to donate, or trade for microscopic prints please pm. Thanks.

yes thats correct, make sure its an internal
bit of flesh 9ie no outside skin attached) buy
the freshest ones you can find.

i normally use tiny pieces as you
only need a little, some times you can split the stem
in half, the peel out strips (the grain will run up the stem)
and place them on agar + h2o2, after a day or
2 you will see the flesh on the agar fluff up with
fresh growth, this wil then move onto the plates.

i dont think it works out too well to rehydrate dryed
shrooms, better the fresher they are.

how many strips, and how long should the strips be, i.e. half the dish long, a quarter long?

tiny pieces seem to be better, as to large and the flesh may rot.
i use bits as thick as cotton, and about 1/5 cm long, 3
of these spaced out on one plate.

break a few oysters up and try to remove inside strips
before you have the agar ready, practise makes perfect.

yeah lol, but bigger will work fine mosty.
I used to use larger chunks, but moved to smaller as i found it better, less flesh, less chance of something attached to the
flesh i.e bacteria.

cool, so mycofile's h202 tek is the way to go huh? I have a glovebox, if I can call it that (rubbermaid with holes cut in it) but is the h202 a better route for an agar newbie?

But I'm sure its fine. If cloning, h2o2 can
really help you out as a newbie, or as a pro.

It will also set you up to getting to a stage
were you can start spores, and do selective
zoneing to develope your own strain :]

Just as an experiment, and to give you an idea
of how good h2o2 is in agar, do a plate without
and try to clone on it. This will show you just
how clean your setup is.

Youll be amazed at what grows on a clean un h202 plate lol.

Paids pretty much spot on as usual but I'd advise against cutting through the mushroom with a sterile scalpel as this could drag contams from the outside into the inner tissue that you are attempting to clone ! I find it best to just break the mushroom apart with your fingers and then clone from the freshly exposed tissue.

you would be better off going to a farmer's market to get the freshest mushrooms to work with. hopefully there is one nearby you.

if you are still confused about the size of tissue to use for cloning, just make sure it is no bigger than the hole in a lifesaver, but preferably smaller.

awesome, thanks for the imagery, only types I could find were shitake, button, and crimini. Too cold for the shitake here. What substrate is best for the other two? Are the oysters still easier than those? I will try to find a good fresh farmers market around, hopefully they will have some.

Oysters are easyer than buttons, shiitake are
also easyer than buttons and all can be grown indoors, so
outside temps shouldnt matter to much.

are the substrates for those species more complicated than the oysters? I like the simplicity of straw. It's easy to get too

Well, not really as you use straw for buttons too, but this
needs to be cased with a bacterialy active casing layer, and
the colonisation time is much longer with buttons.#
Buttons have been cultivated for a long time, though
not for as long as shiitake, so all the shop stock
ones are do-erbal, just oysters make the best starting block.

Clone all you can get though, i did and do, then you
have them and can look into what you need to fruit them :]

For me i preffer that taste of oysters, so I always have
some on the go, that is an important factor, grow
what you like :]

when you grow oysters just beware that they do have the possibility of fruitless reproduction... they can produce something very simmilar to "mushroom piss" but it is apperently laden with spores... also, the ones that I first played with on straw with some grain grew some large rubbery cloud looking things, but never fruited.

I don't remember exactly, but i think it was TMC that I got the info about the asexual reproduction thing.. at any rate, I thought mine were contaminated so I never bothered to help them fruit... (I was growing in a tupperware... EXTREME newbie at the time, before I even knew about the shroomery!)

I have never come across any mention of Pleurotus producing spore laden mushroom piss.
I was not aware that any of the commonly cultivated Pleurotus species could produce spores asexually.
Could you reference "when you grow oysters just beware that they do have the possibility of fruitless reproduction... "

It seems to me that the "simplicity" of straw is not what it's cracked up to be.  Are you going to use high pressure to sterilize stuff, or are you hoping to just pasteurize yer straw?  it's nothing to me, but straw is, uhm, the devil.  no really, i got several species of oyster off on a random mixture of coffee grinds, store bought wood chips for smoking, oat bran, and pelletized aspen from the pet store. 
oysters are easy. 
green stuff is REAL easy.


cloning, on the other hand, is also relatively easy.  cloned oyster from the wild.  in a glove box.  dipped the slightly-larger-than-the hole-in-the-lifesaver piece of flesh into h202/agua and voila. 
i may have even cloned directly to grain from store bought maitake.  but if i did, i wouldn't tell you guys...

page 106 of Growing gourmet and medicinal mushrooms, a great little picture of pleurotus cystidiosus... "This mushroom species is dimorphic - having an alternative life cycle path. The black droplets are resplendent with spores and are not contaminants."

anyway, the same applies for the rest of the pleurotus aswell... it dosen't allways happen, but it is freaquent. my venture would be that these form when conditions are not suitable for full fruitbody production. (now that I think about it and checked, stamets agrees.)

The yellow "mushroom piss", just for clarification is not laden with spores. It is a secondary metabolite, that among other things is paralytic to nematodes. The dark black droplet produced by P. cystidiosus are completely different, both functionally and morphalogically from the the "mushroom piss" which you speak of.

I have a few plates filled with this mushroom piss. Are they still good. They may be a little old I guess. I dont realy want them so if they are ok. I will give them away.

I have a wild clone of P. Ostreatus that I took over a month ago. My petris are definately overincubated. Is there a risk that they have sporulated and have contam'ed my substrain?

I think you will be fine YidakiMan. i too have over-incubated plates of ostreatus but they were still uncontaminated and viable, if a little slower growing. I have never heard of P. ostreatus making spore laden piss.

i am aware that some Pleurotus do form a type of conidia but I think that "anyway, the same applies for the rest of the pleurotus aswell" is a bit reckless and misleading. I have never seen P. pulmonarius or P. ostreatus form any sort of conidia but they will often pin on agar. these are very different things.

Im having truble cloning the store bought oysters, the myc wont grow on my H202 PDA petri. Please read This thread and give me some advice......I really need some help here dudes, its been 10 days and there is still no growth on the oyster plates....

Give it a bit more time mate. Some oysters take off straight away on agar while some don't. The last wild oyster I cloned onto peroxidated PSYA took a couple of weeks to get adjusted to the media.
